Source of material
A mixture of bis-esters (1 mmol) and hydrazine hydrate (0.8 mL, 16 mmol) in absolute ethanol (50 mL) was refluxed for 6 h. The excess solvent was evaporated under reduced pressure to dryness, the obtained residue was triturated with ethanol and the resulting solid was purified from AcOH/H2O to give the bis-hydrazide derivative in 85% yield, mp. 246–248 °C; IR (KBr, cm−1): ν 3470–3218 (NH, NH2), 1678, 1672(2C = O, amide); 1H-NMR (DMSO-d6): δ 1.35 (s, 12H, 4 CH3), 4.15 (s, 4H, 2NH2 D2O exchangeable), 8.18–8.26 (m, 3H, pyridine-H), 8.18, 8.98(2s, 4H, 4NH exchangeable with D2O); 13C-NMR: 25.86(4C, 4CH3), 59.64(2C, NH—C(CH3)2CO), 123.42, 139.00, 149.88(5C, pyridine-C), 160.08, 178.65(4C, 4C = O); MS, m/z (%): 365 (M+, 4), 333(5), 233(100), 148(65), 133(48), 105(56), 77(76) [3].
Experimental details
H atoms were placed in calculated positions and were included in the refinement using the riding model approximation, with Uiso(H) = 1.2Ueq(C). The H atoms of the methyl group were allowed to rotate with a fixed angle around the C—C bond to best fit the experimental electron density (HFIX 137 in the SHELX program suite [2]), with Uiso(H) = 1.5Ueq(C). Nitrogen-bound H atoms were refined subject to distance restraints with Uiso(H) = 1.2Ueq(N).
Discussion
Hydrazines and their analogues constitute a significant group of compounds that has found broad interest in organic synthesis and medicinal chemistry. Several hydrazide–hydrazone derivatives have shown antimicrobial, antiviral, antimalarial, anticonvulsant, antiinflammatory, antituberculosis, antitumor, antiplatelet aggregation, antileishmanial, analgesic and antimalarial properties [4], [5], [6], [7], [8], [9], [10], [11], [12]. Furthermore, various substituted pyridines were recorded to exhibit antibacterial, anti-inflammatory and antitumor activity [13], [14], [15].
In the title compound, the asymmetric unit contains only one independent molecule. The molecules are packed in the crystal structure via six classical intermolecular hydrogen bonds, N3—H1N3⋯O3i, N6—H1N6⋯O2ii, N6—H1N6⋯N4ii, N4—H4B⋯O1iii, N4—H4C⋯O3i, N7—H7A⋯O2iv, Symmetry codes: (i) − x + 1/2, y, z-1/2; (ii) x-1/2, − y + 1, z + 1/2; (iii) − x + 1, − y + 1/2, z-1/2; (iv) − x + 1, − y + 1, z. In addition, the structural is stabilized by several C–H⋯O interactions.
