Tea saponins, extracted from a Camellia oleifera cake, were found to have a potent effect on de-alcoholic activity. To obtain highly pure tea saponins, which can better maintain the activity of alcohol dehydrogenase (ADH), this paper presents an extraction method for tea saponins using deionized water as the extraction agent and a two-stage precipitation method, including ethanol precipitation and CaO precipitation. The optimum conditions for ethanol precipitation were 95% alcohol, a duration of 1.5h and a solid/liquid ratio of 1:4; while the optimum conditions for CaO precipitation were a duration of 2h and an NH 4 HCO 3 /CaO ratio of 2:1. Under the optimum conditions, the content of saponins was 87.58%. The results showed that the greater the amount of tea saponins and the higher its purity, the more significant its activating effect on ADH. When the purity of tea saponins was above 75%, it activated ADH. It indicated that the de-alcoholic mechanism of tea saponins is associated with the activity of ADH. Furthermore, the study characterized the structure of tea saponins by UV absorption and Fourier Transform Infrared (FTIR) spectrometry and LC-MS.

Objective The aim of this study was to investigate the correlation between runt-related transcription factor 3 (RUNX3) gene promoter hypermethylation and gastric cancer risk by meta-analysis. Methods By searching Medline, PubMed, Embase, Cochrane, Ovid and CNKI electronic databases, the open published studies about the association between RUNX3 gene promoter hypermethylation and gastric cancer risk were screened. The hypermethylation rate in cancer tissue and autologous control tissue (normal gastric tissue of gastric cancer patients) were extracted from each included study. The odds ratio (OR) and corresponding 95% confidence interval (95% CI) of RUNX3 gene promoter hypermethylation in cancer tissue versus autologous control tissue of gastric cancer patients were pooled with random or fixed effect models. The publication bias was evaluated by Begg’s funnel plot and Egger’s line regression test. Results Finally, twenty three relevant studies were included in this meta-analysis. The hypermethylation rate in cancer tissue and autologous control tissue of gastric cancer patients were 0.56±0.16 and 0.18±0.22 respectively, which demonstrated a hypermethylation rate in cancer tissue significantly higher than that of autologous controls (P<0.05). A significant positive correlation of hypermethylation rate between cancer tissue and autologous control existed for the included 23 studies(r pearson =0.62, P<0.05). For significant heterogeneity across the studies, the OR was pooled by random effects model. The combined OR was 8.06 with the 95% CI of 5.73~11.32, which indicated the hypermethylation frequency in cancer tissue was higher than that of autologous controls. Conclusion The RUNX3 gene promoter hypermethylation rate was much higher in cancer tissue than that of normal gastric tissue in patients with gastric cancer, which indicates a close association between gastric cancer and RUNX3 gene promoter hypermethylation. Furthermore, RUNX3 gene promoter hypermethylation may be a potential biomarker for gastric cancer diagnosis.

Venous thromboembolism (VTE) refers to the formation of a blood clot inside veins and has a high risk of inducing medical accidents. An effective risk assessment model will help screen high risk populations and prevent the occurrence of VTE. In this study, 287 VTE cases were collected and analyzed for risk factors in a Chinese population. The risks of VTE were evaluated using the Caprini and Padua models. Our results indicated that the Caprini model was more effective in evaluating VTE risk among hospitalized patients than the Padua model. As well, the Caprini model was more relevant in VTE risk assessment among surgery patients compared with internal medicine patients, while the Padua model showed no significant differences. In our studies, the most frequent risk factors included obesity, medical patients currently at bed rest, and severe lung disease. Our studies provide clinical support on selecting the suitable risk assessment model of VTE in the Chinese population.

Objective To investigate the diagnostic value of dual-energy lung perfusion imaging (DEPI) using a dual-source CT system for the pulmonary embolism (PE). Methods 50 patients in high acute PE prevalence were enrolled to accept the DEPI (lung perfusion image and CTA image of pulmonary artery acquired through the Dual Energy software) and emergent DSA angiography (golden diagnostic criterion). Results Patients using CT had significantly reduced examination duration and dosage of contrast agent than those using DSA examination, ( P < 0 . 05 ). In total, 260 pulmonary arteries and 1020 pulmonary segments were examined through CTA, in which embolisms were identified in 50 lobes of lung, 108 pulmonary segments and 82 sub-segments. Reduction or lack of perfusion was identified through DEPI in 48 lobes of lung (concordance rate of 96.0%), 103 pulmonary segments (concordance rate of 95.4%) and 78 subsegments (concordance rate of 95.1%). The comparison of embolism quantity and morphological characteristics of pulmonary artery between CTA images and DEPI images showed no statistically significant difference. Conclusion Better application value can be achieved in the diagnosis of pulmonary embolism by dual-energy lung perfusion imaging using a dual-source CT system.

Agriculture, food industry, and manufacturing are just some of the areas where anaerobic technology can be used. Currently, anaerobic technologies are mainly used for wastewater treatment, solid waste treatment, or for the production of electrical and thermal energy from energy crops processing. However, a clear trend is towards more intensive use of this technology in biomass and biodegradable waste processing and hydrogen or biomethane production. An enormous number of anaerobic digesters are operating worldwide but there is very little information about the effect of different substrate combinations on the methanogens community. This is due to the fact that each of the anaerobic digesters has its own unique microbial community. For the most effective management of anaerobic processes it would be important to know the composition of a consortium of anaerobic microorganisms present in anaerobic digesters processing different input combinations of raw material. This paper characterizes the effect of the input raw materials on the diversity of the methanogen community. Two predominant microorganisms in anaerobic digesters were found to be 99% identity by the sequences of the 16S rRNA gene to the Methanoculleus and Thermogymnomonas genera deposited in GenBank.

CD8 + T cells are a vital component of the adaptive immune system and important for eliminating intracellular pathogens. Notably, mTOR activity is associated with CD8 + T effector memory (Tem) cell differentiation in fungal infections. This study investigates the molecular mechanisms of CD8 + Tem cell proliferation and differentiation mediated by the mTOR pathway in immunosuppressed mice with invasive pulmonary aspergillosis (IPA). We first established the immunosuppressed IPA mouse model, then mice were subjected to rapamycin treatment daily or interleukin (IL)-12 treatment every other day. Lung tissues and blood samples were obtained seven days later. Aspergillus fumigatus was cultured from the lung tissue of mice inoculated with A . fumigatus spores. After IL-12 treatment, the expression of mTOR and its downstream signaling molecule S6 kinase, number of CD8 + Tem cells and interferon-γ expression were significantly increased, while they were significantly decreased after treatment with rapamycin. Additionally, IL-12 treatment induced T-bet but inhibited Eomesodermin expression, while the opposite was seen when the mTOR pathway was blocked by rapamycin. In conclusion, we found that the mTOR pathway induced CD8 + T cell proliferation and differentiation by regulating T-bet and Eomesodermin expression, which significantly influenced immune regulation during IPA and enhanced the immune response against fungal infection.

Effects on seed germination characteristics of 17 tree species were investigated under elevated temperature and CO 2 . Seeds of 5 needle-leaf and 12 broad-leaf species were germinated under four conditions: 24°C + 400 μmol CO 2 mol air –1 , 24°C + 750 μmol CO 2 mol air –1 , 27°C + 400 μmol CO 2 mol air –1 , and 27°C + 750 μmol CO 2 mol air –1 . The elevated temperature and CO 2 affected germination percent (GP) of 7 tree species seeds.GPs of Pinus densiflora , P . thunbergii , Betula ermanii , and Maackia amurensis seeds were affected by the elevated temperature, while only that of P . jezoensis seed was influenced by the elevated CO 2 . GPs of Malus baccata and Zelkova serrata seeds were influenced by both the elevated temperature and CO 2 . In addition, the elevated temperature and CO 2 also affected mean germination time (MGT) of 12 tree species seeds. Particularly, MGTs of P . thunbergii and Rhododendron tschonoskii seeds were influenced by both factors. In conclusion, elevated temperature and CO 2 affected seed germination characteristics, which were reflected by significant differences among tree species. Specifically, these two factors exerted stronger influence on germination pattern such as MGT rather than seed germination percent.

This study is aimed at determining the content of water soluble vitamins in amateur sportsmen before and after the use of reduction diet. Twenty adult male amateur sportsmen aged between 20 and 43 qualified for this study. The participants adhered to individually adjusted reduction diets for 3 months. Diet supplements were excluded from the diet during that time. Vitamins were analyzed before and after the reduction diet using HPLC. The following vitamins were analyzed: B1, B2, B3, PP, B5, B6, B7, B9, B12 and vitamin C. Statistically significant increase of riboflavin in blood plasma (p=0.0254) and statistically significant (p=0.048) decrease in the concentration of niacin was found when compared to the period before the diet. Appropriate level of riboflavin can be obtained by the consumption of proper amount of diary in the diet. Supplementation with PP is advised and, in case of improper diet, the supplementation of B1, B7 and B12 may also be considered.

Background Epigenetics has been recognized as a significant regulator in many diseases. White adipose tissue (WAT) epigenetic dysregulation is associated with systemic insulin resistance (IR). The aim of this study was to survey the differential methylation of genes in obese women with systemic insulin resistance by DNA methylation microarray. Methods The genome-wide methylation profile of systemic insulin resistant obese women was obtained from Gene Expression Omnibus database. After data preprocessing, differing methylation patterns between insulin resistant and sensitive obese women were identified by Student’s t-test and methylation value differences. Network analysis was then performed to reveal co-regulated genes of differentially methylated genes. Functional analysis was also implemented to reveal the underlying biological processes related to systemic insulin resistance in obese women. Results Relative to insulin sensitive obese women, we initially screened 10,874 differentially methylated CpGs, including 7402 hyper-methylated sites and 6073 hypo-methylated CpGs. Our analysis identified 4 significantly differentially methylated genes, including SMYD3 , UST , BCL11A , and BAI3 . Network and functional analyses found that these differentially methylated genes were mainly involved in chondroitin and dermatan sulfate biosynthetic processes. Conclusion Based on our study, we propose several epigenetic biomarkers that may be related to obesity-associated insulin resistance. Our results provide new insights into the epigenetic regulation of disease etiology and also identify novel targets for insulin resistance treatment in obese women.

Background This case-control study investigated an association between breast milk jaundice (BMJ) and infants’ gut microbiome. The study included determination of the diversity of the gut microbiome and identification of bacterial genera associated with BMJ. Methods The study population consisted of 12 infants with BMJ and 22 breastfed infants without jaundice (control). DNA collected from feces was analyzed by PCR amplification and 1% agarose gel electrophoresis, and then sequenced with a MiSeq system. Relative quantification bioinformatics was employed to analyze the DNA sequencing data. An Illumina high-throughput sequencing platform was used to analyze 16S rRNA variable (V) regions V3 and V4 in stool samples. Results In the control group, the proportion of Escherichia/Shigella (genus level) in the gut microbiome (64.67%) was significantly higher than that of the BMJ group. However, the prevalence of Bifidobacterium or Enterococcus in the gut microbiome of the two groups was similar. The Simpson index indicated that the diversity of the bacterial population in the BMJ infants was significantly narrower than in the normal infants. Conclusion The prevalence of Escherichia / Shigella in the gut of breastfed infants is important for lowering BMJ development.

Haemophilus parasuis is an important bacterium affecting pigs, causing Glässer’s disease. To further characterize this species, we determined the complete genomic sequence of H. parasuis CL120103, which was isolated from diseased pigs. The strain H. parasuis CL120103 was identified as serovar 2. The size of the largest scaffold is 2,326,318 bp and contains 145 large contigs, with the N50 contig being 20,573 bp in length. The complete genome of H. parasuis CL120103 is 2,305,354 bp in length with 39.97% GC content and contains 2227 protein-coding genes, 19 ribosomal rRNA operons and 60 tRNA genes. Sequence similarity of the genome of H. parasuis CL120103 to the previously sequenced genome of H. parasuis was up to 96% and query cover to 86%. Annotation of the genome of H. parasuis CL120103 identified a number of genes encoding potential virulence factors. These virulence factors are involved in metabolism, adhesion, secretion and LPS biosynthesis. These related genes pave the way to better understand mechanisms underlying metabolic capabilities. The comprehensive genetic and phylogenetic analysis shows that H. parasuis is closely related to Actinobacillus pleuropneumoniae and provides a foundation for future experimental confirmation of the virulence and pathogen-host interactions in H. parasuis .

Bakcground We measured expression of miRNA-145 in gallbladder carcinoma and its influence on propagation, invasion, and apoptosis of gallbladder carcinoma cells in vitro. Methods miRNA-145 expression was compared between normal gallbladder epithelial cells and GBS-SD (gallbladder series) cells using miRNA chip technology. Propagation, apoptosis, and invasion properties of each cell group were tested using MTT, a clone-formation assay, flow cytometry, Western blot, and Transwell assays. Results Expression of miRNA-145 was observed to be down-regulated and GBC-SD cell clones transiently transfected with hsa-miRNA-145 were substantially reduced compared with controls (p<0.01). We observed that GBC-SD cells transfected with hsa-miRNA-145 and double-positive (Annexin V and PI) for apoptosis were more numerous than controls. Moreover, GBC-SD cells over-expressing miRNA-145 had significantly greater expression of apoptosis-related protein, caspase-3. A Transwell assay confirmed that GBC-SD cells over-expressing miRNA-145 that migrated to the lower chamber were fewer compared with controls. Post-transcriptional regulation of gene expression was measured using dualluciferase reporter assays and data show that miRNA-145 facilitates the inhibition of GBC-SD cell growth and invasion while inducing apoptosis by targeting DFF45. Conclusion Thus, we speculate that miRNA-145 facilitates inhibition of GBC-SD cell growth and invasion while inducing apoptosis by targeting DFF45; however, miRNA-145 does not directly affect the GBC-SD cell cycle.

Osteosarcoma (OS) is a commonly occurring primary malignant bone cancer with serious impact and high mortality, yet effective and safe therapy method not available. The aim of the present study was to elucidate the antitumor effect of telmisartan on human osteosarcoma cells in vitro and its underlying mechanism. The proliferation effect of osteosarcoma cell lines U2OS was examined by Cell Counting Kit-8. The invasive and migratory capabilities were determined by transwell invasion and migration assay. The percentage of apoptotic cells were detected by flow cytometric analysis and proteins related to apoptosis including Bax, Bcl-2 and Cleaved Caspase-3 were examined by western blotting. The expressions of mammalian target of rapamycin (mTOR) signaling relevant molecules were detected by western blot assay. Telmisartan treatment caused dose-dependent and time-dependent inhibition of proliferation and inducing anti-migration, anti-invasiveness and apoptosis of U2OS cells. The induction of apoptosis was confirmed concurring with the altered expression of proteins associated with the apoptosis. Mechanistically, telmisartan suppresses mTOR activation. Telmisartan can impede the growth, invasion, migration and induce the apoptosis of U2OS cell probably through inhibiting the mTOR signaling pathway activation. Thus, telmisartan is a potential drug for the prevention and treatment of human osteosarcomal cancer.

The main purpose of our study is to optimize the formulation for Ginkgolide B (GB) solid dispersion in order to improve its dissolution in water. For the preparation of GB solid dispersion, we use a solvent method. The optimized formulation consists of GB : PVPK30 = 1:10, PVPK30 as the carrier, and ethanol : dichloromethane = 1:1 as the solvent. They were treated ultrasonically for 10 min at 60°C. The results from scanning electron microscopy (SEM), differential scanning calorimetry (DSC), X-ray diffraction (XRD), and particle size analysis show that the morphological appearance of GB changed significantly in solid dispersion. The original crystal form of GB no longer existed, but it was uniformly dispersed within PVPK30 in a non-crystalline form. It is probably because the -C=O in GB forms hydrogen bonds with the -OH of PVPK30 or urea; this produces a nice solid dispersion and significantly improves the dissolution of GB in water. When GB is in a solid dispersion system, GB’s dissolution in water could be enhanced from 30% to 80%. Furthermore, it may even be produced as a solid agent.

This study was conducted to determine the potential nutritive value and in vitro gas production (IVGP) parameters of Olea europaea L . (Olive = OL ) , Morus alba L . (Mulberry = ML) and Citrus aurantium L . (Sour orange = SOL) tree leaves. Hohenheim gas test was used to determine the in vitro gas productions of the leaves. The gas production of samples over time was recorded for 3, 6, 9, 12, 24, 48, 72 and 96 h after incubation. Completely Randomized Design was used to compare gas production, and gas production kinetics of samples. The findings of the present study suggested that there were differences among the tree leaves in terms of crude protein, NDF, in vitro gas productions, organic matter digestibility (OMD), metabolisable energy (ME), net energy lactation (NE L ) and relative feed values (RFV) (P<0.01). ML had the highest condensed tannin contents (P<0.05), in vitro gas production (IVGP), OMD and energy values (P<0.01). SOL had highest RFV values. OL showed the lowest IVGP when compared to SOL and ML. Low NDF and ADF contents of SOL would probably increase the voluntary intake, digestibility and relative feed values of these leaves by ruminants. In conclusion, it was determined that OL, ML and SOL used in the study have low in vitro gas production and can be utilized as alternative roughage feed in ruminants. However, it is recommended that the results obtained from this research should be tested in in vivo studies.

Most studies highlight mesenchymal stem cells (MSCs) extracted primarily from bone marrow (BM), very few report the use of peripheral blood (PB), often due to the associated low seeding density and difficulties with extraction techniques. As ageing populations are becoming more predominant globally, together with escalating demands for MSC transplantation and tissue regeneration, obtaining quality MSCs suitable for induced differentiation and biological therapies becomes increasingly important. In this study, BM and PB were obtained from elderly patients and extracted MSCs grown in vitro to determine their successful isolation and expansion. Patients’ socio-demographic background and other medical information were obtained from medical records. Successful and failed cultures were correlated with key demographic and medical parameters. A total of 112 samples (BM or PB) were used for this study. Of these, 50 samples (44.6%) were successfully cultured according to standardised criteria with no signs of contamination. Our comparative analyses demonstrated no statistical correlation between successful MSC cultures and any of the six demographic or medical parameters examined, including sample quantity, age, sex, race, habits and underlying comorbidities of sample donors. In conclusion, the present study demonstrates that typical demographics and comorbidities do not influence successful MSC isolation and expansion in culture.

Background To analyze the application value of apparent diffusion coefficient (ADC) and exponent apparent diffusion coefficient (EADC) in evaluating the efficacy of radiation and chemotherapy in cervical cancer using pre- and posttreatment diffusion-weighted magnetic resonance imaging (DW-MRI) scans. Methods 52 patients with cervical cancer were administered radiation and chemotherapy. Both MRI and DW-MRI were obtained at different stages. The ADC and EADC values, as well as the maximum tumor diameter, were measured and analyzed. Results We found that the ADC value increased after treatment, and the EADC value decreased. Changes in the calculated ADC occurred earlier than the morphologic changes of the tumors. A negative correlation was detected between reduced rates in the maximum tumor diameter two months after treatment and pretreatment ADC value (r = –0.658, P < 0.05). An ROC curve and nonlinear regression analysis showed that the formula, y = (1525500.122x 2 – 4689.962x + 3.482) × 100%, can be used to calculate the percentage of complete remission after treatment according to the pretreatment ADC value. Conclusion Our data suggest that pretreatment ADC and EADC values are predictive of the efficacy of radiation and chemotherapy. Both ADC and EADC values during treatment were instrumental in early monitoring and dynamic observation.

The influence of stress factors on a plant may lead to worse functioning of the plant and the loss of its crop. The effect of Aulacorthum solani feeding on Trollius europaeus with regard to active compounds in the leaves was investigated in the study. The antioxidant activity of the leaves, the material on which the insects fed, was compared with that of the material that was not infested by the aphids. Stress level was evaluated on the basis of such physiological parameters as chlorophyll fluorescence and photosynthesis activity. An increase of 34.5% in the content of polyphenolic compounds, as compared to control, was demonstrated in the material exposed to the biotic stress caused by aphids. The content of phenolic acids was 28% higher while that of flavonoids rose by 25%. The increase in polyphenolic compounds augmented the antioxidant activity of the material.

The effectiveness of Pl genes is known to be resistant to downy mildew (DM) disease affected by fungus Plasmopara halstedii in sunflower. In this study phenotypic analysis was performed using inoculation tests and genotypic analysis were carried out with three DM resistance genes Plarg, Pl13 and Pl8 . A total of 69 simple sequence repeat markers and 241 F 2 individuals derived from a cross of RHA-419 (R) x P6LC (S), RHA-419 (R) x CL (S), RHA-419 (R) x OL (S), RHA419 (R) x 9758R (S), HA-R5 (R) x P6LC (S) and HA89 (R) x P6LC (S) parental lines were used to identify resistant hybrids in sunflower. Results of SSR analysis using markers linked with downy mildew resistance genes ( Plarg , Pl8 and Pl13 ) and downy mildew inoculation tests were evaluated together and ORS716 (for Plarg and Pl13), HA4011 (for Pl8 ) markers showed positive correlation with their phenotypic results. These results suggest that these markers are associated with DM resistance and they can be used successfully in marker-assisted selection for sunflower breeding programs specific for downy mildew resistance.

Objective Given the role of basic fibroblastic growth factor (bFGF) in tumor growth, it has been considered as a potential target for tumor therapy. In this study, we investigate the effect of bFGF antagonistic peptide on the growth and angiogenesis of 4T1 mammary carcinoma tumor (MCT) in BALB/c mice. Methods An engineered peptide was injected into BALB/c mice in doses of 1, 2.5, 5 and 10 mg/kg daily for 14 days. Immunohistochemical analysis using anti-CD31 and anti-CD34 were conducted as indices of angiogenesis. In addition, blood samples were taken from the eyes of treated and control mice and the levels of Interleukin-8 (IL-8) and Tumor Necrosis Factor-α (TNF-α) were measured by ELISA. Data was analyzed by ANOVA using SPSS. Results The antagonistic peptide inhibited growth and angiogenesis of MCT ( P ≤0.05), and decreased the serum level of IL-8 and TNF-α in treated groups compared to the control groups. Conclusion The inhibition of tumor angiogenesis has been considered as an important strategy to halt tumor growth. The results of current study confirm that the antiangiogenic peptide effectively inhibited the growth of MCT, and shows potential for clinical trials for the treatment of cancer in humans.

This study investigated the antihyperglycemic and antihyperlipidemic effects of low-molecular-weight carrageenan (LC) on rats fed a high-fat diet. Wistar rats were divided into five groups: normal control group (NC), high-fat diet control group (HC), carrageenan-treated control group (CC), 1% LC group (1% LC), and 3% LC-groups (3% LC). Body weight, food intake, fecal weight, blood glucose, and serum lipid levels were measured. After 30 days, body weight significantly decreased in the LC-treated groups than in the HC group. Moreover, in the LC-treated groups, postprandial blood glucose, total cholesterol, triglyceride, and low-density lipoprotein cholesterol (LDL-C) levels decreased, whereas high-density lipoprotein cholesterol (HDL-C) levels increased. From this study, our data suggest that LC has antihyperglycemic and hypolipidemic effects when compared to carrageenan, likely related to its increased absorption due to its lower molecular weight.

The use of rivers for recreational and domestic practices makes it imperative to scrutinize the water quality circulating within surrounding communities. The complexity of biological, physical and chemical constituents in water is constantly evolving. This study evaluated various microbial and physico-chemical parameters in a polluted river system over a 12-month period. Apart from an increase in chemical pollutants, elevated levels of E. coli , total (TC) and faecal (FC) coliforms, and Shigella species could be attributed to faecal contamination entering the catchment. Canonical correspondence analysis revealed a strong relationship between FC, TC and temperature whereas moderate interactions was seen between total dissolved solids, electrical conductivity, TC and FC populations. Furthermore, close relationships between the bacterial and phage communities were also observed. The complex interactions of these physico-chemical and microbial indicators could be due to anthropogenic activities, changing climatic conditions and the excreta of infected and non-infected individuals entering the river. Assessing the complexity of aquatic ecosystems can aid in the development of novel, customizable, inexpensive water purification tools.

Objective This study is aimed at exploring the role of TUFT1 in osteosarcomas. Methods We investigated the expression of TUFT1 in osteosarcoma cell lines and explored the correlation between TUFT1 expression and prognosis in osteosarcoma patients based on the expression data downloaded from Gene Expression Omnibus (GEO) website. The effects of TUFT1 on osteosarcoma cell proliferation, migration and invasion were investigated by silencing TUFT1 in osteosarcoma MG63 cell line. Finally, western blot was performed to determine the expression changes of MAPK signaling pathway related proteins after silencing TUFT1. Results We found that the expression of TUFT1 was significantly up-regulated in osteosarcoma cell lines compared with the normal control. Using Kaplan-Meier analysis, we identified that high TUFT1 expression was positively correlated with poor prognosis in osteosarcoma patients. Furthermore, knockdown of TUFT1 remarkably inhibited MG63 cell proliferation, migration and invasion. Using western blot analysis, we found that the phosphorylation levels of MEK and ERK were reduced obviously in MG63 cells after silencing TUFT1 (p<0.01). Conclusions Our results demonstrated that TUFT1 plays a promoting role in MG63 cell proliferation and metastasis and has the potential to be a predictor as well as a therapeutic target for osteosarcoma patients.

Background Human bone marrow mesenchymal stem cells (BMSCs) are of great significance for bone regeneration and bone formation. Long non-coding RNAs (lncRNAs) may be involved in modulating cell differentiation. This study aimed to investigate the role of lncR-2271 in promoting osteogenic differentiation in human BMSCs. Methods Human BMSCs were infected using lncR-2271 overexpression (group A) with lentiviral system or transfected with lncR-2271 siRNA (group B). Cells transfected with scrambled plasmids were used as a negative control (group C). Osteogenesis markers were evaluated using alkaline phosphatase (ALP) activity, RUNX2 and osterix (OSX) at protein levels and calcification by Alizarin Red staining. Results BMSCs from group A showed significantly higher ALP activity compared to BMSCs in group B and control group (group C) at both days 7 and 14 following osteogenic induction; ALP activity was significantly lower in the group B compared to the group C. RUNX2 and OSX protein expressions were significantly higher in group A and significantly lower in group B, compared to those in group C, respectively. At day 21, calcification in human BMSCs in group A was significantly higher compared to groups B and C as shown by Alizarin Red staining; calcification was significantly lower in group B compared to group C. Conclusion Our data suggested lncR-2271 played a role in promoting osteogenic differentiation in human BMSCs. This study is the first to illustrate the important role of lncR-2271 in bone formation.

Objective The current study aimed to explore the predictive ability of serum uric acid (SUA) in patients suffering from acute ST segment elevation myocardial infarction (STEMI). Method PubMed, EMBASE, Cochrane Library, and Medline databases were systematically searched from their respective inceptions to February 2018. Systematic analysis and random-effects meta–analysis of prognostic effects were performed to evaluate STEMI outcomes [i.e., in-hospital mortality, one-year mortality, in-hospital Major Adverse Cardiovascular Events (MACE)] in relation to SUA. Results A total of 12 studies (containing 7,735 patients with acute STEMI) were identified (5,562 low SUA patients and 3,173 high SUA patients). Systematic analysis of these studies showed that high SUA patients exhibited a higher incidence of in-hospital MACE (OR, 2.30; P < 0.00001), in-hospital mortality (OR, 3.03; P < 0.0001), and one-year mortality (OR, 2.58; P < 0.00001), compared with low SUA patients. Conclusions Acute STEMI patients with high SUA exhibited an elevated incidence rate of in-hospital MACE, in-hospital mortality, and one-year mortality. Further randomized controlled trials will be needed to verify these results.

Resource sharing between the connected ramets of clonal plants through physiological integration can increase the tolerance of plants to environmental stress. However, the role of physiological integration in the translocation of heavy-metal pollutants between different habitats receives little attention, especially in the aquatic-terrestrial ecotones. An amphibious clonal plant Alternanthera philoxeroides was used to simulate plant expansion from unpolluted soil to a chromium (Cr)-polluted water environment. Basal older ramets growing in unpolluted soil were connected or disconnected with apical younger ramets of the same fragments in polluted environments at different Cr concentrations. Harvested basal ramets were also used for decomposition tests for the loss of residual mass and release of Cr to soil. With increasing Cr concentration there was reduction in biomass of the apical ramets, especially those separated from the basal parts. Cr was detected in the basal ramets with connection to apical parts. The decomposition of plant litter from the basal ramets connected with polluted apical parts might release retained Cr to unpolluted soil. The amount and chemical forms of Cr in the plant litter changed over time. It is concluded that Cr could be transferred from polluted aquatic to unpolluted terrestrial habitats through amphibious clonal plants.

The purpose of the present investigation is to examine the function of the C2H2-type zinc finger transcription factor of Arabidopsis thaliana 6 ( ZAT6 ) in salt stress tolerance in cells of rice ( Oryza sativa L.), cotton ( Gossypium hirsutum L.) and slash pine ( Pinus elliottii Engelm.). Cells of O. sativa , G. hirsutum , and P. elliottii overexpressing ZAT6 were generated using Agrobacterium -mediated genetic transformation. Molecular and functional analysis of transgenic cell lines demonstrate that overexpression of ZAT6 increased tolerance to salt stress by decreasing lipid peroxidation and increasing the content of abscisic acid (ABA) and GA 8 , as well as enhancing the activities of antioxidant enzymes such as ascorbate peroxidise (APOX), catalase (CAT), glutathione reductase (GR), and superoxide dismutase (SOD). In rice cells, ZAT6 also increased expression of Ca 2+ -dependent protein kinase genes OsCPK 9 and OsCPK 25 by 5–7 fold under NaCl stress. Altogether, our results suggest that overexpression of ZAT6 enhanced salt stress tolerance by increasing antioxidant enzyme activity, hormone content and expression of Ca 2+ -dependent protein kinase in transgenic cell lines of different plant species.

Aim Sini decoction (SND) is a commonly used herbal formula showing lipid-lowering effects and is applied in traditional Chinese medicine (TCM) for the treatment of cardiovascular disease (CVD) and atherosclerosis (AS). However, the mechanisms behind its anti-atherosclerotic effects are still unknown, and will be investigated in this study. Methods AS was induced in rabbits by high fat diet (HFD) and treated with solvent (HFD group), atorvastatin (Ator group), or SND (SND group) for 12 weeks. Healthy rabbits (Chow group) were used as control. Serum and liver homogenates were collected, and lipid profiles as well as serum ApoA-I and ApoB were examined. Histopathological changes and lipid deposition in the proximal aorta and liver were detected by Oil red O staining. Western blot was used to detect the expression of ABCA1, ApoA-I, ApoB, PPARγ, and LXRα in liver, peritoneal macrophages, peripheral mononuclear cells (PMC), and adipose tissues. Results SND significantly attenuated the levels of total cholesterol (TC), triglyceride (TG), and low density lipoprotein cholesterol(LDL-C) in serum and liver. However, high density lipoprotein cholesterol (HDL-C) dramatically increased. SND treatment also decreased lipid deposition and improved the structure of the liver and aorta. Furthermore, SND enhanced the expression levels of ABCA1, PPARγ, and LXRα in liver, adipose tissues, PMC, and peritoneal macrophages. It also upregulated hepatic and serum ApoA-I expression and serum ApoA-I/ApoB ratio. CONCLUSIONS:SND treatment relieved AS, improved lipid profiles, and increased serum HDL-C level. The potential mechanism behind this might be the improvement of reverse cholesterol transport (RCT) involved with enhanced expression of ABCA1, ApoA-I, PPARγ, and LXRα.

The timely and accurate diagnosis of ascites is of great significance for early treatment and prognostication. This study explored the value of soluble myeloid triggering receptor expressed on myeloid cell 1 (sTREM-1) and C-reactive protein (CRP) for assessing ascites. A total of 133 patients with ascites who received treatment at the Affiliated Hospital of Taishan Medical University between September 2015 and September 2017 were retrospectively analyzed. The ascites in 22, 45, 33 and 33 patients were tuberculous, bacterial, tumorous, and transudative, respectively. Healthy volunteers (n=30) who received a health examination at the same hospital during the same period constituted the control group. Before treatment, both ascitic sTREM-1 and CRP showed significant differences among the ascites subgroups (P<0.001), with the highest levels in the bacterial subgroup. Serum sTREM-1 and CRP also showed significant differences among the groups. A correlation analysis showed a positive correlation between sTREM-1 and CRP. ROC curves of the bacterial subgroup showed that when the optimal cutoff point was set to 20.2, the sensitivity, specificity, positive predictive value, and negative predictive value of the serum sTREM-1 index were 0.933, 0.955, 0.914, and 0.965, respectively. sTREM-1 may provide more diagnostic value than CRP for the diagnosis of bacterial ascites.

A simple separation method was used in this study to directly separate laccase-producing fungi from withered plant materials. A laccase-producing filamentous fungus was isolated and purified. The strain was highly similar to the species in genus Trametes by ITS sequence analysis, and therefore named Trametes sp. MA-X01. The addition of cupric ions and aromatic compounds to the liquid medium could induce the laccase synthesis in Trametes sp. MA-X01. Copper-induced laccase activity increased in a dose-dependent manner. The highest laccase activity (2138.9 ± 340.2 U/L) was obtained by adding 2.5 mM Cu 2+ to the culture medium, which was about 7 times higher than that of the control group. The induction degree of aromatic compounds was different. For the present study, the highest laccase activities were obtained by adding vanillic acid (1007.9±59.5 U/L) or vanillin (981.6±77.2 U/L) to the medium, which were 3.5 and 3.4 times higher than the laccase activity of the control group, respectively.

Hydrolytic enzyme production by thermophilic bacteria isolated from hot springs in the southern region of Saudi Arabia was investigated. The physical and chemical properties of the hot springs prove to be an important environment for hydrolytic-enzyme-producing thermophilic bacteria. Eighty-four bacterial isolates were obtained from three hot springs: Al-Majardah, Al-Khubah and Al-Ardah. Screening of the isolates for enzyme production indicated that 78 isolates showed activity for one or more enzymes. Molecular identification and phylogenic analysis of selected promising isolates confirmed the identity of the isolates as Bacillus aerius, Bacillus licheniformis and Bacillus sonorensis , which have potential to produce the target enzymes α-amylase, protease and lipase, respectively. Optimization of hydrolytic enzyme production by bacterial strains was investigated using kitchen waste as a cheap carbon energy source. Maximum enzyme production was achieved after 72 hours of incubation at the beginning of the stationary phase of growth. Enzyme production was dependent on the initial pH value in the range of pH 7.5-8.5 and an optimal incubation temperature of between 55-60°C. Enzyme production increased gradually in proportion to the kitchen waste concentration; whereas maximum lipase production was gained at 5.0% (w/v) kitchen waste, 7.0% (w/v) of waste was optimal for both α-amylase and protease productivity. The results indicated that hot springs in Saudi Arabia are a substantial source of thermophilic bacteria producing industrially important enzymes using cheap and unexploited waste.

Intestinal sulfate-reducing bacteria are often isolated from patients with inflammatory bowel disease, including ulcerative colitis, and can be involved in the development of gut inflammation. A comparison of the metabolism of intestinal sulfate-reducing bacteria isolated from individuals with colitis and healthy controls using statistical analysis has never been studied and described before. The aim of our research was to evaluate the parameters of dissimilatory sulfate reduction in Desulfovibrio species that were isolated from the feces of healthy objects and individuals with colitis. Principal component analysis indicates that the strains that were isolated from individuals with colitis grouped in the same cluster by biomass accumulation and sulfide production, same as the strains isolated from healthy individuals. Sulfate and lactate consumption measured over time showed negative correlation (Pearson correlations, p <0.01), healthy: -0.760; colitis: -0.770; healthy: -0.828; colitis: -0.847, respectively. The calculated linear regression (R 2 ) was lower in biomass accumulation and hydrogen sulfide production, 0.531; 0.625 respectively. Thus, biomass accumulation and sulfide production, together with measured kinetic parameters play an important factor in bowel inflammation, including ulcerative colitis. Additionally, acetate production can also synergize with H 2 S, while sulfate consumption and lactate oxidation likely represent minor factors in bowel disease.

Emodin, a major component of rhubarb, has anti-tumor effects in a variety of cancers, influencing multiple steps of tumor development through modulating several signaling pathways. The aim of this study is to examine the effect of emodin on cell apoptosis and explore the underlying mechanisms in human endometrial cancer cells. Here we report that emodin can inhibit KLE cell proliferation and induce apoptosis in a time- and dose-dependent manner. Western blot assay found that emodin was involved in MAPK and PI3K/Akt signaling pathways. Specifically, emodin significantly suppressed the phosphorylation of AKT, and enhanced the phosphorylation of MAPK pathways. Furthermore, the generation of reactive oxygen species (ROS) was up-regulated in KLE cells upon treatment with emodin, while the anti-oxidant agent N-acetyl cysteine (NAC) can inhibit emodin-induced apoptosis and promote the activation of AKT and Bcl-2. Taken together, we revealed that emodin may induce apoptosis in KLE cells through regulating the PI3K/AKT and MAPK signaling pathways, indicating the importance of emodin as an anti-tumor agent.

It has been demonstrated that miR-539 plays an important role in the development and progression of tumors. The purpose of this study was to analyze the correlation between the expression level of miR-539 and the clinicopathological features and prognosis of patients with pancreatic cancer. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to analyze the expression level of miR-539 in 60 patients with pancreatic cancer. It was found that miR-539 gene expression was down-regulated in pancreatic cancer compared with that in paracancerous tissues. In addition, the expression level of miR-539 was inversely correlated with tumor differentiation (poorly to moderately differentiated vs . well differentiated, P =0.006), lymph node metastasis (positive vs . negative, P =0.006), clinical stage (III-IV vs . I-II, P =0.002), CA199 (≥200 vs . <200, P =0.019) and distant metastasis (positive vs . negative, P =0.035). The survival time of pancreatic cancer patients with low expression of miR-539 was significantly shorter than that of patients with high expression of miR-539. Multivariate analysis suggested that miR-539 expression level was an independent prognostic indicator for patients with pancreatic cancer ( P =0.025). Down-regulation of miR-539 may be a potentially unfavorable prognostic factor for patients with pancreatic cancer, and further studies are needed to confirm our conclusion in the future.

Fascioliasis is a disease of livestock which is now recognized as an emerging disease in humans. Cantharellus cibarius and Ganoderma applanatum are known for their medicinal properties. The use of ethanolic extracts of these macrofungi against the eggs and miracidia of Fasciola spp. is a promising method to break the parasite transmission cycle. The aim of the study is to evaluate the inhibitory effects of ethanolic extracts of the mushrooms on eggs and miracidia of Fasciola spp. Concentrated eggs and miracidia of Fasciola spp. were exposed to different concentrations (1-8 mg/ml) of extracts of Ganoderma applanatum (GEE) and Cantharellus cibarius (CEE) at different time intervals. GEE showed superior antiparasitic activities when compared to CEE at all concentrations tested. Significant positive correlations were observed between the concentration of GEE and mortality in miracidia (r=0.980, P <0.05) and CEE and mortality in miracidia (r= 0.968, P <0.05). The study showed that ethanolic extracts of G. applanatum and C. cibarius have ovicidal and miracicidal activities. While G. applanatum showed excellent activities, activities in C. cibarius were moderate. Therefore, these mushroom extracts can be regarded as promising sources of bioactive compounds that could be developed into ovicides and miracicides.

Objective This study aimed to evaluate the expression of progestin and adipoQ receptor family member VI (PAQR6, mPRδ) in prostate cancer and to explore its role in prostate cancer progression. Methods PAQR6 mRNA expression was evaluated based on the data obtained from the TCGA database and the GEO database. The prognostic value of PAQR6 was explored by Kaplan-Meier analysis. To investigate the role of PAQR6, it was depleted by siRNA in DU145 cells. The effects of depleting PAQR6 on DU145 cell viability and migration were determined by CCK8 assay, colony formation assay, and wound healing assay, respectively. The activation of MEK and ERK were analyzed by western blot. Results PAQR6 mRNA expression was significantly up-regulated in prostate cancer tissues and correlated with lower survival rates (p=0.014). Furthermore, qPCR revealed that PAQR6 expression was elevated in DU145 and LNCaP cells compared with RWPE-2 cells. Depleting PAQR6 obviously suppressed DU145 cell proliferation and migration (p<0.01). In addition, the ratio of p-MEK/MEK and p-ERK/ERK was significantly reduced after silencing PAQR6 (p<0.01). Conclusion PAQR6 might play a facilitating role in prostate cancer development by regulating the MAPK signaling pathway. Moreover, it might serve as a potential predictor and therapeutic target in prostate cancer.

NAC (for N AM , A TAF1-2 , and C UC2 ) proteins are one of the largest transcription factor families in plants. They have various functions and are closely related to developmental processes of fruits. Tomato ( Solanum lycopersicum ) is a model plant for studies of fruit growth patterns. In this study, the functional characteristics and action mechanisms of a new NAC-type transcription factor, SlNAC3 (SGN-U568609), were examined to determine its role in tomato development and ripening. The SlNAC3 protein was produced by prokaryotic expression and used to immunize New Zealand white rabbits to obtain a specific polyclonal antibody against SlNAC3. By co-immunoprecipitation and MALDI-TOF-MS assays, we showed that there was an interaction between the SlNAC3 protein and Polygalacturonase-2 (PG-2), which is related to the ripening and softening of fruit. Chromatin immunoprecipitation assays revealed the genome of the highly stress-tolerant Solanum pennellii chromosome 10 (sequence ID, HG975449.1), further demonstrating that SlNAC3 is a negative regulator of drought and salinity stress resistance in tomato, consistent with previous reports. These results indicate that SlNAC3 is not only involved in abiotic stress, but also plays a necessary role in mediating tomato ripening.

Objective The aim of this study was to evaluate the expression of Angiopoietin-1 (Ang-1), Angiopoietin-2 (Ang-2) and vascular endothelial growth factor (VEGF) in cervical cancer and its clinical significance. Methods Immunohistochemical assay was used to examine the expression of Ang-1/2 and VEGF in tumor tissue from 56 cervical squamous cell carcinoma patients treated with operation only (SCC-O group), as well as 51 subjects with cervical squamous cell carcinoma treated with neoadjuvant radiotherapy (SCC-RCO group, n=28) or neoadjuvant chemotherapy (SCC-CO group, n=23). Both microvessel density (MVD) and lymphatic vessel density (LVD) were examined in the three groups through detection of CD34 and D2-40 expression in respective tissue samples. Results With the progression of cervical cancer, the positive expression scores of Ang-2 and VEGF were significantly increased (p<0.05). Compared with surgical intervention, neoadjuvant chemoradiotherapy significantly reduced the positive expression scores of Ang-1, Ang-2, and VEGF in cervical cancer tissues (p<0.05). The MVD values of the SCC-CO and SCC-RO groups were significantly reduced as compared to the SCC-O group (p<0.05). Similarly, the LVD values of the SCC-CO and SCC-RO groups were also significantly reduced when compared to those of the SCC-O group (p<0.05). However, LVD values of the SCC-CO and SCC-RO groups were not statistical different (p>0.05). Conclusion Ang-1, Ang-2 and VEGF may play an important role in the development of cervical cancer. Mutual synergism of Ang-2 and VEGF demonstrated a close relationship with the generation of cervical blood and lymphatic vessels. Cervical cancer radiotherapy and chemotherapy could significantly inhibit the formation of blood vessels and lymphatic vessels in tumor tissue.

Autologous skin grafts are used to treat severe burn wounds, however, the availability of adequate donor sites makes this option less practical. Recently, stem cells have been used successfully in tissue engineering and in regenerative medicine. The current study aims to differentiate umbilical cord tissue derived mesenchymal stem cells (CT-MSCs) into skin cells (fibroblasts and keratinocytes) for use to treat severe burn wounds. After isolation, MSCs were characterized and their growth characteristics were determined. The cells were induced to differentiate into fibroblasts and keratinocytes using respective induction medium. Results indicated that CT-MSCs were spindle shaped, plastic adherent and positive for CD29, CD44, CD73, CD90 markers. CT-MSCs also showed high proliferative potential as indicated by cumulative population doubling, doubling time and plating efficiency. The MSCs were successfully differentiated into fibroblast and keratinocytes as indicated by morphological changes and expression of lineage specific genes. We propose that these differentiated skin cells which are derived from CT-MSCs can thus be used for the development of bioengineered skin; however, further studies are required to evaluate the utility of these substitutes.

Enterococcus faecium , is an important nosocomial pathogen with increased incidence of multidrug resistance (MDR) – specifically Vancomycin resistance. E. faecium constitutes the normal microbiota of the human intestine as well as exists in the hospitals and sewage, thus making the microorganism difficult to eliminate. Phage therapy has gained attention for controlling bacterial MDR infections and contaminations. We have successfully isolated from waste water and characterized a lytic bacteriophage STH1 capable of targeting Vancomycin resistant Enterococcus faecium (VREF) with high specificity. The phage was isolated from sewage water of a hospital at district Dera Ismail Khan, Pakistan. Initial characterization showed that magnesium and calcium ions significantly increased phage adsorption to the host. One step growth experiment showed a latent period of 18 min with burst size of 334 virions per cell. Optimal temperature and pH of the phage was 37°C and 7.0, respectively. Phage application to host strain grown in milk and water (treated and untreated) showed that the phage efficiently controlled bacterial growth. The study suggests that the phage STH1 can serve as potential control agent for E. faecium infections in medical facilities and in other environmental contaminations.

Anthracyline (ANT) has been demonstrated as a useful treatment for leukemia and solid tumors. However, ANT has previously reported cardiotoxic effects, which can reduce the therapeutic index for cancer treatment. This study aimed to investigate the associations of glycogen phosphorylase isoenzyme BB (GPBB), myoglobin (Mb), and brain natriuretic peptide (BNP) with anthracycline (ANT-induced cardiotoxicity (AIC)) amongst the Chinese population. Patients suffering from leukemia were recruited. Electrocardiogram and echocardiography were used along with chemotherapy to determine left ventricular ejection fraction (LVEF), mitral ratio of peak early to late diastolic filling velocity (E/A), E-wave deceleration time (EDT), and isovolumic relaxation time (IVRT). Double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) was employed to examine and compare serum GPBB, Mb, and BNP levels. Following chemotherapy, the patients presented higher levels of serum GPBB, Mb, and BNP than before chemotherapy treatment. The levels of LVEF (%), E/A, and IVRT were significantly decreased after chemotherapy, while EDT was markedly increased. The cumulative ANT dose was positively corelated to serum GPBB, Mb, and BNP levels while it was negatively corelated to LVEF levels. In conclusion, serum GPBB, Mb, and BNP levels in combination might provide higher diagnostic accuracy in the early detection of AIC compared with other single indicators.

Previous studies have suggested that Bcl2-associated athanogene 2 (BAG2) serves as a crucial regulator for tumorigenesis in multiple tumors. However, little is known about the effect of BAG2 on esophageal squamous cell carcinoma (ESCC). This study focused on investigating whether BAG2 functions as a cancer-promoting gene in ESCC. In this work, gene expression data and clinical information from the NCBI Gene Expression Omnibus (GEO), Oncomine and The Cancer Genome Atlas (TCGA) were collected and analyzed. Expression of BAG2 in ESCC was determined using quantitative reverse transcription polymerase chain reaction (qRT-PCR). BAG2 was knocked down using small interference RNA (si-RNA) approach. Cell proliferation, migration and invasion were assessed by Cell Counting Kit-8 (CCK-8) and transwell assays. Molecular mechanism was detected by western blotting assay. The expression of BAG2 both in ESCC tissues and cells was upregulated and overexpression was associated with worsened prognosis. BAG2 silencing inhibited ESCC cell proliferation, migration and invasion, which was regulated by the phosphatidylinositol-3-kinase (PI3K)/ protein kinase B (AKT) signaling pathway. These results reveal contributions of BAG2 as a predictor and potential therapeutic target in ESCC.

We investigated climatic trends in two contrasting locations in Europe at a regional level and at two specific sites, and we analysed how these trends are associated with the dry matter yield (DMY) of agriculturally improved grasslands. Trends of different meteorological variables were evaluated for Wielkopolska province, central Poland (1985-2014) and Troms county, northern Norway (1989-2015), as well as for two research stations located in these regions. Significant trends of increased mean air temperatures annually, and in April, June, July, August and November were identified both at the regional and site levels in Wielkopolska. In addition, growing degree days were increasing in Wielkopolska. In Troms, the common trends for the region and site studied were increase in mean air temperature in May and decrease in January. Grassland DMY was subsequently regressed against those meteorological variables for which significant trends were detected. In the Wielkopolska region, yields were negatively associated with the increase in air temperature in June, August, and the annual air temperature. The last relationship was also detected at the site level. We did not find any significant effects of climate trends on grassland DMY in the Norwegian study site or region.

Halophytes are plants that exhibit high salt tolerance, allowing them to survive and thrive under extremely saline conditions. The study of halophytes advances our understanding about the important adaptations that are required for survival in high salinity conditions, including secretion of salt through the salt glands, regulation of cellular ion homeostasis and osmotic pressure, detoxification of reactive oxygen species, and alterations in membrane composition. To explore the mechanisms that contribute to tolerance to salt stress, salt-responsive genes have been isolated from halophytes and expressed in non-salt tolerant plants using targeted transgenic technologies. In this review, we discuss the mechanisms that underpin salt tolerance in different halophytes.

This work describes the current state of research on the potential relationship between protein content in human saliva and dental caries, which remains among the most common oral diseases and causes irreversible damage in the oral cavity. An understanding the whole saliva proteome in the oral cavity could serve as a prerequisite to obtaining insight into the etiology of tooth decay at early stages. To date, however, there is no comprehensive evidence showing that salivary proteins could serve as potential indicators for the early diagnosis of the risk factors causing dental caries. Therefore, proteomics indicates the promising direction of future investigations of such factors, including diagnosis and thus prevention in dental therapy.

Nanoparticles have many applications both in industry and medicine. Depending upon their physical and chemical properties, they can be used as carriers of therapeutic molecules or as therapeutics. Nanoparticles are made of synthetic or natural polymers, lipids or metals. Their use allows for faster transport to the place of action, thus prolonging its presence in the body and limiting side effects. In addition, the use of such a drug delivery system protects the drug from rapid disintegration and elimination from the body. In recent years, the use of proteins and peptides as therapeutic molecules has grown significantly. Unfortunately, proteins are subject to enzymatic digestion and can cause unwanted immune response beyond therapeutic action. The use of drug carriers can minimize undesirable side effects and reduce the dose of medication needed to achieve the therapeutic effect. The current study presents the use of several selected drug delivery systems for the delivery of proteins, peptides and other therapeutic molecules.

Organisms have evolved a spectrum of strategies that facilitate survival in the face of adverse environmental conditions. In order to make full use of the unfavorable resources of nature, human beings usually impose selective pressures to breed phenotypic traits that can survive in adverse environments. Animals are frequently under attack by biotic stress, such as bacterial and viral infections, while plants are more often subjected to abiotic stress, including high salinity, drought, and cold. In response to these diverse stresses, animals and plants initiate wide-ranging changes in gene expression by altering regulation of transcriptional and post-transcriptional activities. Recent studies have identified a number of key responsive components that promote survival of animals and plants in response to biotic and abiotic stresses. Importantly, with recent developments in genome-editing technology based on the CRISPR/Cas9 system, manipulation of genetic elements to generate stress-resistant animals and plants has become both feasible and cost-effective. Herein, we review important mechanisms that govern the response of organisms to biotic and abiotic stresses with the aim of applying our understanding to the agriculture and animal husbandry industries.

In rapid proliferating cancer cells, there is a need for fast ATP and lactate production, therefore cancer cells turn off oxidative phosphorylation and turn on the so called "Warburg effect". This regulating the expression of genes involved in glycolysis. According to many studies, glucose transporter 1, which supplies glucose to the cell, is the most abundantly expressed transporter in cancer cells. Hexokinase 2, is one of four hexokinase isoenzymes, is also another highly expressed enzyme in cancer cells and it functions to enhance the glycolytic rate. The up-regulation of these two proteins has been established as an important factor in promoting development and metastasis in many types of cancer. Furthermore, other enzymes involved in glycolysis pathway such as phosphoglucose isomerase and glyceraldehyde 3-phosphate dehydrogenase, exhibit additional functions in promoting tumor growth in a non-glycolytic way. This review demonstrates the pivotal role of GLUT1, HK2, PGI and GAPDH in cancer development. In particular, we look at how the multifunctional proteins, PGI and GAPDH, affect cancer cell survival. We also present various clinical cancer cases in terms of the overexpression of selected proteins, which may be considered as a therapeutic target.

Zenia insignis Chun is a large, fast-growing deciduous tree. In this study, we successfully developed a reliable and efficient protocol for the regeneration of fertile plants via callus induction from leaf segments of young Z. insignis seedlings. The best results were obtained with a medium containing 11.00 μM 6-benzyladenine (6-BA), 1.20 μM indole-3-butytric acid (IBA), and 0.45 μM 2,4-dichlorophenoxyacetic acid (2,4-D), which yielded morphogenic callus within 2 weeks at a frequency of 62.23%. We tested the effect of IBA alone and in combination with 6-BA on the bud differentiation response of Z. insignis callus. Shoots differentiated normally when cultured on differentiation medium containing 6.00 μM 6-BA and 1.20 μM IBA. Regenerated buds elongated successfully in medium containing 1.20 μM gibberellic acid (GA3). The elongated shoots were then transferred to Murashige and Skoog basal medium supplemented with various combinations of naphthalene acetic acid (NAA) for root induction; well-developed roots were achieved on MS basal medium supplemented with 0.01 μM NAA at a rooting rate of 89.23%. Rooted plantlets were successfully acclimatised to a greenhouse at a survival rate exceeding 90.00%.

Herbal medicinal products (HMPs) have grown increasingly popular in the United States, many of them with imported raw materials and sold online. Yet due to the lack of regulation from the US Food and Drug Administration (FDA), manufacturers of the products can substitute or add in other herbs that are not advertised on the label. In this study, as part of the Urban Barcode Research Program (UBRP), an education initiative to engage New York City high school students in science, we aimed to taxonomically authenticate single-ingredient online-sold HMPs containing non-native plants through DNA barcoding of the internal transcribed spacer 2 region (ITS2) and matK . We were able to successfully barcode 20 HMPs, but four of these did not match the expected species. It was concluded that the four HMPs advertising astragalus, epazote, ginseng, and chanca piedra were contaminated/ substituted because their ITS2 and matK DNA sequences did not match the expected taxonomy in GenBank, a government database. Our study highlights the importance of herbal pharmacovigilance in the absence of strict government regulation of herbal supplements and motivates crowd-sourced DNA barcoding to enable American consumers make informed choices and be more empowered to safeguard their health.

Background The clinical course of myasthenia gravis (MG) during pregnancy is highly variable and unpredictable. The management of MG in pregnancy has not been standardized. Methods Eight cases of MG in pregnancy, who were treated and gave birth in our hospital between 2004 and 2012, were retrospectively reviewed. Results In three patients, MG deteriorated during pregnancy. Three patients discontinued their medication for MG during their pregnancy, and the other five patients continued on corticosteroid or pyridostigmine. None of the infants showed any congenital abnormalities. Interestingly, there was a trend towards lower birth weight in infants born to women who had an exacerbation of MG during pregnancy. One patient who had unstable MG before pregnancy and voluntarily discontinued the medication for MG at the beginning of pregnancy, experienced MG exacerbation at the 30th week of pregnancy and gave birth prematurely to an infant with transient neonatal MG at the 34th week. The other seven patients had uneventful full-term pregnancy. Conclusion Women with unstable MG should postpone pregnancy to avoid potential risk of MG exacerbation and adverse effects on the fetus. Medication for MG should not be stopped abruptly during pregnancy, particularly for women with unstable MG. MG during pregnancy should be closely monitored and properly controlled.

Object To summarize the etiology, pathology, diagnosis, clinical features, and treatment of the rare extranodal Rosai-Dorfman disease (RDD). Methods Clinical data of three cases of extranodal RDD who were admitted to the E. N. T. Department of the second hospital of Jilin University were analyzed retrospectively, and the literature was reviewed. Results Three cases of extranodal RDD (maxillary sinus, nasal pharyngeal focus, and external auditory meatus) had a low fever and weight loss rather than painless enlarged lymph nodes. Surgical intervention was managed to clarify the diagnosis. All pathological diagnoses were extranodal RDD. Predisone for oral use was given to all patients postoperatively. There was no recurrence in the following 3 months, except case 1 was lost 2 months later. Conclusions RDD is a rare idiopathic histiocytic proliferative disorder defined by its unique histopathological features: a proliferation of huge histiocytic cells with emperipolesis and S-100(+), CD1a(-). RDD is characterized clinically by bilateral cervical painless enlarged lymph nodes, while extranodal RDD is rarer and its manifestations varied. A defined therapeutic regimen has not been elucidated. RDD in about 20% of patients is self-limited. Surgical intervention is the main management of treatment, with glucocorticoids used in initial medical therapy. More clinical trials are necessary before drawing conclusions.

Chlamydiosis is an important zoonosis which can transmit from birds to humans, and investigation first reported the seroprevalence of Chlamydia psittaci in black - headed gulls ( Larus ridibundus ) at the Dianchi Lake, China. A total of 1029 serum samples collected from black - headed gulls between 2012-2015 were analyzed. The gulls were randomly caught and blood collected at Dianchi Lake, China. All the samples were analyzed for the presence of antibodies to C . psittaci by indirect hemagglutination assay (IHA). In this survey, the total infection rate was 11.86% (122/1029). The results of the present survey documented the existence of relatively high C . psittaci seroprevalence in black-headed gulls, which have a potential risk to the wild bird health and human health. Comprehensive practical control approaches and measures should be executed.

The essential oil of Mosla chinensis Maxim cv. Jiangxiangru is known for its antibacterial ability. This study aimed to investigate the chemical composition of Jiangxiangru essential oil and its inhibitory effect on Staphylococcus aureus biofilm formation. Gas chromatography/mass spectrometry (GC–MS) was used to determine the chemical composition of Jiangxiangru essential oil. Subsequently, the eight major chemical components were quantitatively analyzed using GC– MS, and their minimum inhibitory concentration (MIC) values against S . aureus were tested. Biofilm formation was detected by crystal violet semi-quantitative method and silver staining. Of the 59 peaks detected, 29 were identified by GC–MS. Of these peaks, thymol, carvacrol, p -cymene, γ-terpinene, thymol acetate, α-caryophyllene, 3-carene, and carvacryl acetate were present at a relatively higher concentration. The results of the quantitative test showed that thymol, carvacrol, p -cymene, and γ-terpinene were the major components of the essential oil. Among the eight reference substances, only thymol, carvacrol, and thymol acetate had lower MICs compared with the essential oil. Essential oil, carvacrol, carvacryl acetate, α-caryophyllene, and 3-carene showed the better inhibition of S . aureus biofilm formation. When one fourth of the MIC concentrations were used for these substances (0.0625 mg/mL for essential oil, 0.0305 mg/mL for carvacrol, 1.458 mg/mL for carvacryl acetate, 0.1268 mg/mL for α-caryophyllene, and 2.5975 mg/mL for 3-carene), the inhibition rates were over 80%. However, thymol, γ-terpinene, thymol acetate, and p -cymene showed a relatively poor inhibition of S . aureus biofilm formation. When 1× MIC concentrations of these substances were used, the inhibition rates were less than 50%. In conclusion, Jiangxiangru essential oil and its major components, carvacrol, carvacryl acetate, α-caryophyllene, and 3-carene, strongly inhibited biofilm formation in S . aureus .

Polar microbial derived antibiotics have potential as alternatives to traditional antibiotics in treating fish against pathogenic bacteria. In this paper, 23 strains of polar fungi were fermented to detect bacteriostatic products on three aquatic pathogenic bacteria, subsequently the active fungus was identified. It was indicated that secondary metabolites of 23 strains weredistinct; of these, the extract of strain B-7 (belonging to Bjerkandera according to molecular identification) demonstrated a strong antibacterial activity to Streptococcus agalactiae , Vibrio anguillarum and Aeromonas hydrophila ATCC7966 by Kirby-Bauerpaper strip method. During one fermentation cycle, the pH curve of the fermentation liquor became lowest (4.0) on the 4 th day and rose back to 7.6 finally after 5 days, The residual sugar curve was decreased before stablising on the 6 th day. It is presumed that a large amount of alkaline secondary metabolites might have been produced during fermentation. This study focuses on antagonism between aquatic pathogenic bacteria and fermentation metabolites from Antarctic fungi for the first time, which may provide data on research of antibiotics against aquatic pathogenic bacteria.

Aim The aim of the study is to investigate a variation in the gene SLC6A19 in a female patient with Hartnup disorder manifested only by seizure. Methods DNA samples collected from the patient and her parents were analyzed and twelve exons of the SLC6A19 gene were amplified and sequenced. Results We found c.47C>T and c.1522G>A mutations in the gene SLC6A19 belonging to the patient, which are missense mutations inherited from her parents. The c.47C>T mutation is from her father and c.1522G>A is inherited from her mother. The parents are both heterozygous healthy carriers. Conclusion Two novel mutations of the SLC6A19 gene are revealed in the female patient with Hartnup disorder, exhibiting no typical dermatologic problems, but having dramatic neurological symptoms.

The normal megastrobilli and microstrobilli before and after the sexual reversal in Pinus massoniana Lamb. were studied and classified using a transcriptomic approach. In the analysis, a total of 190,023 unigenes were obtained with an average length of 595 bp. The annotated unigenes were divided into 56 functional groups and 130 metabolic pathways involved in the physiological and biochemical processes related to ribosome biogenesis, carbon metabolism, and amino acid biosynthesis. Analysis revealed 4,758 differentially expressed genes (DEGs) between the mega- and microstrobili from the polycone twig. The DEGs between the mega- and microstrobili from the normal twig were 5,550. In the polycone twig, 1,188 DEGs were identified between the microstrobili and the sexually reversed megastrobili. Concerning plant hormone signal transduction pathways, the DEGs from both the normal and polycone twigs displayed distinct male or female associated expression patterns. There were 36 common hormone-related DEGs from the two types of twigs of P . massoniana . Interestingly, expression of these DEGs was up-regulated in the bisexual strobili, which underwent the sexual reversal. A portion of MADS-box genes in the bisexual strobili were up-regulated relative to expression in microstrobili.

Endoplasmic reticulum stress is one of the mechanisms of cell apoptosis. In this study, the mechanism of oxymatrine-induced human esophageal cancer Eca-109 cell apoptosis by the endoplasmic reticulum stress pathway was investigated. Eca-109 cells were cultured in vitro with different doses of oxymatrine (0.5, 1, 2 μg/mL) for 48 h. The cell viability and proliferation inhibition rate were examined by MTT assay and cell cycle assay. The apoptosis rate was examined by Annexin V-FITC/propidium iodide assay. The expression of endoplasmic reticulum stress markers, including binding immunoglobulin protein and CCAAT-enhancer-binding protein homologous protein, were determined by real-time quantitative polymerase chain reaction and western blotting, respectively. MTT data showed that oxymatrine significantly inhibited the proliferation of Eca-109 cells. The cell apoptosis rate was quantified by flow cytometry. The expression of binding immunoglobulin protein was markedly downregulated in oxymatrine-treated Eca-109 cells while that of CCAAT-enhancer-binding protein homologous protein was upregulated. Oxymatrine inhibited proliferation and induced apoptosis of human esophageal carcinoma Eca-109 cells. Thus, oxymatrine may be a potential agent for treating human esophageal cancer.

Skewed sex development is prevalent in fish hybrids. However, the histological observation and molecular mechanisms remain elusive. In this study, we showed that the interspecific hybrids of the two fish species, Oreochromis niloticus and Oreochromis aureus , had a male ratio of 98.02%. Microscopic examination revealed that the gonads of both male and female hybrids were developmentally retarded. Compared with the ovaries, the testes of both O. niloticus and hybrids showed higher DNA methylation level in two selected regions in the promoter of cyp19a , the gonadal aromatase gene that converts androgens into estrogens, cyp19a showed higher level gene expression in the ovary than in the testis in both O. niloticus and hybrid tilapia. Methylation and gene expression level of cyp19a were negative correlation between the testis and ovary. Gene transcription was suppressed by the methylation of the cyp19a promoter in vitro. While there is no obvious difference of the methylation level in testis or ovary between O. niloticus and hybrids. Thus, the DNA methylation of the promoter of cyp19a may be an essential component of the sex maintenance, but not a determinant of high male ratio and developmental retardation of gonads in tilapia hybrids.

With the rapid development of information technology and biomedical engineering, people can get more and more information. At the same time, they begin to study how to apply the advanced technology in biomedical information. The main research of this paper is to optimize the machine learning method by particle swarm optimization (PSO) and apply it in the classification of biomedical data. In order to improve the performance of the classification model, we compared the different inertia weight strategies and mutation strategies and their combinations with PSO, and obtained the best inertia weight strategy without mutation, the best mutation strategy without inertia weight and the best combination of the two. Then, we used the three PSO algorithms to optimize the parameters of support vector machine in the classification of biomedical data. We found that the PSO algorithm with the combination of inertia weight and mutation strategy and the inertia weight strategy that we proposed could improve the classification accuracy. This study has an important reference value for the prediction of clinical diseases.

We report the case of a thirty-eight-year-old woman admitted to our hospital due to palpitation and chest distress. ST-T segment change was found in her ECG. She was then diagnosed with hypertrophic cardiomyopathy by two-dimensional echocardiography. Physical examination showed no obvious abnormal signs and all laboratory examinations were within the normal range. Myocardial fibrosis was detected by cardiac magnetic resonance imaging (MRI). A novel heterozygous mutation (c.235C>T/p.Arg79Cys) in TNNI3 for cardiac troponin I was identified in her. Subsequently, her families were investigated. No one died suddenly in her family. Her father, one of her siblings and one of her daughters had the same genetic mutation but with different clinical manifestations while the others were healthy. Her father and brother were also diagnosed with hypertrophic cardiomyopathy with different clinical manifestation. However, the echocardiography of her daughter was absolutely normal. We hypothesized that the Arg79Cys mutation in TNNI3 leads to a slow development of cardiac hypertrophy and the phenotype of this gene mutation is diverse.

The aim of this study is to assess the winter wheat planting (WWP) area in Henan Province and investigate its temporal and spatial variations by using remote sensing (RS) technology. A spectral angle mapper (SAM) was adopted to identify the WWP area of each district divided by the hierarchical grades of land surface drought index during 2001-2015. The results obtained show the expediency of monitoring the WWP areas at the regional scale via drought regionalization, which provides a goodness-of-fit R 2 =0.933, a mean relative error MRE=49,118 ha, and an overall accuracy up to 90.24%. The major WWP areas in Henan Province were located in Zhoukou, Zhumadian, Shangqiu, Nanyang, and Xinxiang prefecture-level cities. Two representative sites are mountainous districts, with rich water resources or high urbanization rate, which have a low probability of WWP. Both sites exhibited a strongly manifested evolution of WWP areas, which could be attributed to extremely cold weather conditions, crop alternation, the popularization of new varieties, and fast expansion of built-up areas. The results of this study are instrumental in the analysis of crop planting variation characteristics, which should be taken into account in the further decision-making process related to the crop planting strategies.

This study was undertaken to evaluate the clinical efficacy of serum soluble triggering receptors expressed by myeloid cell-1 (sTREM-1), procalcitonin (PCT), C-reactive protein (CRP) and lactic acid (Lac) as biomarkers for death risk within 28 days in patients with severe sepsis. Fifty-one cases of severe sepsis from the department of ICU in Lishui People’s Hospital from May 2013 to February 2017 were retrospectively analyzed. These cases were divided into survival (n=39) and death (n=12) groups based on the outcome within 28 days of treatment. Serum levels of sTREM-1, PCT, CRP and Lac were measured on the day of admission and compared between the survival and death groups. And the death prediction value within 28 days were evaluated according to serum sTREM-1, PCT, CRP and Lac. The serum level of TREM-1 and Lac were 128.70±46.10 pg/mL, 7.02±1.56 mmol/L for the death group and 83.69±26.57 pg/mL 4.44±0.45 mmol/L for survival group. The serum levels of sTREM-1 and Lac in death group were significantly higher than those of survival group (p<0.05). However, the serum PCT and CRP between the survival and death group were not statistically different (p>0.05). The death prediction sensitivity, specificity and AUC within 28 days were high for serum sTREM-1 (75.00%, 77.78%, 0.79 ) and APACHEII (74.89%, 84.62%, 0.84). However, the prediction value of serum level PCT, CRP and Lac were relatively low. A significant positive correlation was found between serum sTREM-1 and APACHEII score r pearson =0.54, (p<0.001). However, no such correlation was observed between serum CRP, Lac and APACHEII scores (p>0.05). Conclusion Serum sTREM-1 was significantly elevated in sepsis patients who died within 28 days of admission, suggesting that this test could be a potential biomarker for severe sepsis patients, and also be used for prognostic evaluation.

Objective The purpose of this study was to investigate the correlation between Interleukin 17 (IL-17) gene rs3748067 C>T polymorphism and gastric cancer risk through pooling the open published data. Method Case-control or cohort studies relevant to IL-17 gene rs3748067 C>T polymorphism and gastric cancer susceptibility were systematic searched for in the databases of CNKI, Pubmed, Medline, Embase and Web of science. The association between IL-17 gene rs3748067 C>T polymorphism and gastric cancer risk were expressed with an odds ratio(OR) and 95% confidence interval (95% CI). Statistical heterogeneity across the studies was evaluated by I2 test. Publication bias was evaluated by Begg’s funnel plot and Egger’s line regression test. Results Finally, seven case-control studies were included in our present study. Because of the statistical heterogeneity among the included studies for the aspects of dominant (TT+CT vs CC), recessive (TT vs CT+CC) and homozygous genetic model (TT vs CC), the data was pooled by random effect model. The pooled ORs were OR=0.99 (95% CI: 0.65-1.52), OR =1.23 (95% CI: 0.73-2.06 ) and OR=1.14 (95% CI: 0.58-2.27) for dominant, recessive and homozygous genetic model respectively. The pooled data indicated no correlation between IL-17 gene rs3748067 C>T polymorphism and gastric cancer risk. Significant publication bias was found in the dominant genetic model (p<0.05), but not in recessive and homozygous genetic model (p>0.05). Conclusion Based on the present evidence, there was no correlation between IL-17 gene rs3748067 C>T polymorphism and gastric cancer susceptibility in all genetic model. However, for the small sample size, significant heterogeneity and publication bias, the conclusion should be further evaluated through well designed case-control or cohort studies.

Objective To investigate the efficacy of Danhong injection on the serum concentration of tumor necrosis factor alpha (TNF-α), interleukin 6 (IL-6) and nuclear factor kappa-light-chain-enhancer of activated B (NF-κB) in rats with intracerebral hemorrhage (ICH) and evaluate its therapeutic effects on inflammation and cerebral edema. Methods Sixty male Wistar rats were randomly divided into control, model and Danhong groups with 25 rats in each group. Intracerebral injection of autologous arterial blood was performed on model and Danhong groups in order to establish intracerebral hemorrhage model. Rats in the control group were given the same operation procedure without blood injection. After successfully establishing the intracerebral hemorrhage model, the rats were given Danhong (2ml/kg/d) through intraperitoneal injection. Rats in the control and model groups were given the same amount of normal saline respectively. The brain water content (BWC) and serum level of TNF-α, IL-6 and NF-κB were measured in all groups at the time points of day 1, 3, 5, 7 and 9. Results The neurological deficit score (NDS) were not statistical different in days 1, 3 and 5 between the model and Danhong group (P>0.05); However, on day 7 and 9 after modeling, the NDS in the Danhong group was significant lower than that of the Model group (P<0.05). The brain water content in the model and Danhong groups were significantly elevated compared to control group (P<0.05). The brain water content was significant elevated after modeling in the model and Danhong groups on day 3 and gradually decreased over the next 6 days. The brain water content was significantly higher in the model group for days 3 to 9 compared to the Danhong group (P<0.05). Compared to the model group, the serum NF-κb was significantly lower in the Danhong group for the time point of day 3 and 5 (P<0.05); However, compared to the model group, the serum TNF-α and IL-6 levels in the Danhong group were significantly lower for each time point (P<0.05). Conclusion Danhong injection can reduce cerebral edema in rats with cerebral hemorrhage, and protect the brain nerve function. These effects may be related to its function of regulating serum TNF-α, NF-κB and IL-6 expression.

Objective This work proposes to predict target genes and pathways for uveal melanoma (UM) based on an ensemble method and pathway analyses. Methods: The ensemble method integrated a correlation method (Pearson correlation coefficient, PCC), a causal inference method (IDA) and a regression method (Lasso) utilizing the Borda count election method. Subsequently, to validate the performance of PIL method, comparisons between confirmed database and predicted miRNA targets were performed. Ultimately, pathway enrichment analysis was conducted on target genes in top 1000 miRNA-mRNA interactions to identify target pathways for UM patients. Results: Thirty eight of the predicted interactions were matched with the confirmed interactions, indicating that the ensemble method was a suitable and feasible approach to predict miRNA targets. We obtained 50 seed miRNA-mRNA interactions of UM patients and extracted target genes from these interactions, such as ASPG , BSDC1 and C4BP . The 601 target genes in top 1,000 miRNA-mRNA interactions were enriched in 12 target pathways, of which Phototransduction was the most significant one. Conclusion: The target genes and pathways might provide a new way to reveal the molecular mechanism of UM and give hand for target treatments and preventions of this malignant tumor.

Objective To evaluate the practical effectiveness of smart metal artifact reduction (SMAR) in reducing artifacts caused by metallic implants. Methods Patients with metal implants underwent computed tomography (CT) examinations on high definition CT scanner, and the data were reconstructed with adaptive statistical iterative reconstruction (ASiR) with value weighted to 40% and smart metal artifact reduction (SMAR) technology. The comparison was assessed by both subjective and objective assessment between the two groups of images. In terms of subjective assessment, three radiologists evaluated image quality and assigned a score for visualization of anatomic structures in the critical areas of interest. Objectively, the absolute CT value of the difference (ΔCT) and artifacts index (AI) were adopted in this study for the quantitative assessment of metal artifacts. Results In subjective image quality assessment, three radiologists scored SMAR images higher than 40% ASiR images (P<0.01) and the result suggested that visualization of critical anatomic structures around the region of the metal object was significantly improved by using SMAR compared with 40% ASiR. The ΔCT and AI for quantitative assessment of metal artifacts showed that SMAR appeared to be superior for reducing metal artifacts (P<0.05) and indicated that this technical approach was more effective in improving the quality of CT images. Conclusion A variety of hardware (dental filling, embolization coil, instrumented spine, hip implant, knee implant) are processed with the SMAR algorithm to demonstrate good recovery of soft tissue around the metal. This artifact reduction allows for the clearer visualization of structures hidden underneath.

Objective The aim of the study was to investigate whether nucleophosmin type A mutation ( NPM1A ) in plasma was associated with the prognosis of patients with acute myeloid leukemia (AML). Methods Plasma NPM1A levels were investigated in 80 AML patients, 22 patients with benign hematopathy and 12 healthy donors by qRT-PCR. Additionally, the relationship between NPM1A levels and clinic characteristics were evaluated by Chi-square test. Kaplan-Meier method was used to analyze overall survival (OS) and relapse-free survival (RFS), and univariate and multivariate analyses were performed with Cox proportional hazard model. Results Plasma levels of NPM1A in AML patients were significantly higher than those in benign hematopathy patients and healthy controls, respectively (both P<0.001). Additionally, high NPM1A level was significantly associated with higher WBC and platelet count (both, P<0.05). Moreover, survival analysis revealed that patients with high NPM1A levels had worse OS (P<0.001) and RFS (P<0.001). Multivariate analysis identified NPM1A as an independent prognostic predictor for AML (OS: HR=8.214, 95% CI: 2.974-22.688, P<0.001; RFS: HR=4.640, 95%CI: 1.825-11.795, P=0.001). Conclusions Results reveal that NPM1A in plasma could serve as an ideal tool for predicting the prognosis of patients with AML.

Background Hepatocellular carcinoma is one of the leading causes of cancer-related death. Hepatic transcatheter arterial chemo-embolization (TACE) is commonly used clinically for advanced hepatocellular carcinoma treatment. AIM The aim of this study was to evaluate whether arterial infusion of rapamycin can improve the effect of TACE in treatment of rabbit hepatocellular carcinoma. Material and Methods Eighteen healthy New Zealand white rabbits weighing 2.6 ± 0.3 kg were used in a standardised hepatocellular carcinoma model and randomly divided into three groups of 6 rabbits. Group A: the rabbits were treated with rapamycin and TACE by administering arterial perfusion of 2 mg/kg rapamycin + 1 mg/kg epirubicin, 0.2 mg/kg mitomycin, and lipiodol emulsion embolization. Group B: rapamycin was reduced to 1 mg/kg. And for Group C, the rabbits received only TACE treatment. 14 days post operation, CT scan and digital subtraction angiography (DSA) was performed to examine TACE efficacy. The rabbits were killed by air embolism and the expression of HIF-1a, VEGF, iNOS, and CD34 were measured in an immunohistochemical assay of thetumor tissue. Results HIF-1a, VEGF and iNOS protein expression in Group A was significantly lower than that of Group B and Group C (P<0.05). The tumor MVD in group C was significantly higher than that of group A and group B (P<0.05); and the tumor MVD of group B was significantly higher than group A (P<0.05). Conclusion Arterial infusion of rapamycin combined with TACE can improve treatment efficacy by decreasing HIF-1a, VEGF, iNOS and CD34 expression.

Antiarrhythmic drugs are widely used, however, their efficacy is moderate and they can have serious side effects. Even if catheter ablation is effective for the treatment of atrial fibrillation and ventricular tachycardia, antiarrhythmic drugs are still important tools for the treatment of arrhythmia. Despite efforts, the development of antiarrhythmic drugs is still slow due to the limited understanding of the role of various ionic currents. This review summarizes the new targets and mechanisms of antiarrhythmic drugs.
