1 Source of materials
To a solution of d-tryptophan methyl ester hydrochloride (2.55 g, 10 mmol) in methanol (30 mL) was added 1-naphthaldehyde (1.87 g, 12 mmol). The mixture was refluxed for 3 h, until the TLC indicated the reaction was completed. The solvent was evaporated to dryness, yielding the crude product that was then purified to obtain a single crystal of high quality. For crystal growth, the crude product was dissolved in a minimal amount of hot ethanol and slowly cooled to room temperature.
2 Experimental details
Isotropic displacement factors were applied to the hydrogen atoms with the formula U iso(H) = 1.2 times U eq(C). All hydrogen atoms were refined under the assumption that they are bonded to their respective parent atoms.
3 Comment
The side chain of tryptophan is indole, an aromatic compound with a dual-ring structure. The biosynthesis pathway of tryptophan is known to be the most complex and energy-intensive among all amino acids. 5 Crystal structures of tryptophan derivatives have been extensively reported. 6 – 8
In the crystal structure of the compound under investigation, the C atoms in the naphthalene ring and the C and N atoms in the indole ring are nearly coplanar. The N atom in the piperidine ring is situated in a distinct plane from the other carbon atoms, with a dihedral angle of 43.0(4)° for C12–C11–N2–C15. The angle between the planes of the naphthalene ring and the indole ring is measured at 79.7°. All bond lengths, bond angles, and dihedral angles align with those of previously reported crystal structures. 9 – 11
