The abundance of elk ( Alces alces ) in Poland at the beginning of 2016 reached over 28 000 individual animals. However, little is still known about health status of elk population in the country. In particular, important health issue tend to be parasitic diseases which are the most common diseases in wild animals. Special emphasis should be given to the parasitofauna of elks. Their ability to move over great distances increases the chance of parasitic infestations in new areas and transmission to other wild animals and livestock. The aim of this article is to describe the parasites occurring in elk in Poland on the basis of previous studies from the country and to compare them with data from neighboring countries. The presented review reveals that elk populations have not been examined for the presence of parasites for decades, and no recent studies in Poland have examined the parasitofauna of this animal. As such, our study shows that assessing the parasites present in elk populations remains an important area of research.

Of the three species of Entamoeba found in swine, namely E. histolytica , E. polecki and E. suis , E. histolytica can also infect humans, producing colitis or abscesses of liver and leading to death. However, the pathogenicity of other species of Entamoeba has not been fully characterized. Here, we conducted histopathology and molecular surveys on a pig farm where piglets had blackish feces or muddy diarrhea. Histopathological examination of two piglets showed necrosis of the mucous surface at the ileum, cecum, or colon, infiltration of neutrophils, and formation of ulcers. Based on morphological characteristics, E. polecki and E. suis trophozoites were mainly detected at lamina propria and surface of the lesion, respectively, and Lawsonia intracellularis , a bacterial pathogen, was also detected. Molecular analysis using the small subunit ribosomal RNA gene on other piglets and a sow revealed infection with both E. polecki and E. suis . These findings corroborate our previous reports that the two Entamoeba spp. are pathogenic in pigs as aggravations of symptoms with L. intracellularis . This is the first report about mixed infection with E. polecki and E. suis .

This research was undertaken to study the molecular detection and characterization of Cryptosporidium spp. in environmental water sources at Samsun and Giresun Provinces of The Black Sea in Turkey. Two-hundred forty and one-hundred eighty environmental samples were collected from a total of twenty and twenty-five sampling sites of Giresun and Samsun Provinces. One hundred twenty untreated drinking water samples were also detected for Cryptosporidium spp. in both investigated areas. 101 (%42), 92 (%38.3) of 240 and 74 (41.1%), 70 (38.8%) of 180 environmental samples have been found positive for Cryptosporidium spp. by Loop mediated isothermal amplification (LAMP) targeting the S-adenosyl-L-methionine synthetase (SAM) gene and nested PCR targeting small subunit (SSU)rRNA gene in Samsun and Giresun Provinces, respectively. Of the tested untreated drinking water samples collected from the investigated area, one sample was positive for Cryptosporidium spp. Six and twelve samples were clearly sequenced for the Cryptosporidium (SSU)rRNA gene among the highest positive samples selected from each of the twenty and twenty-five sampling sites of Giresun and Samsun Provinces, respectively. Genetic characterization of Cryptosporidium isolates from water samples represented Cryptosporidium bovis for five samples, Cryptosporidium parvum for six samples and one sample for Cryptosporidium felis in Samsun Province, where C. parvum for five samples and C. bovis for one sample were sequenced in Giresun Province. According to accessible information sources, this is the first research about genotyping of Cryptosporidium spp. in water samples collected from Samsun and Giresun Provinces of Turkey.

Digenetic trematodes have complex life cycles involving multiple hosts and free-living larval stages. Some species have 2 lar-val stages that infect snails, with miracidia and cercariae using these molluscs as first and second intermediate hosts, respec-tively. Although both larval stages may infect the same snail species, this is accomplished using different chemical cues and may be influenced by different biotic and abiotic factors. Significant differences in the infection patterns of these parasitic stages regarding host size and density were observed in 2 separate field studies. The prevalence of sporocysts/rediae and mean abundance of Echinostoma spp. metacercariae infection were positively correlated with host size, while the prevalence of Echinostoma spp. cercariae infection was positively correlated with host density across 5 different pulmonate snail species. Larger snails within a given species tend to be older and the increased exposure time may be responsible for the positive correlations with host size. Additionally, infection by miracidia in more vagile snail hosts was influenced by trematode species richness at a sample site, which may be attributed to increased encounter rate as a result of increased movement by the snail hosts. Echinostoma spp. metacercariae prevalence was influenced by host density, possibly due to high abundances of larval clones and their response to more generalized chemical cues attributed to low host specificity by cercariae. Although they can infect the same gastropod hosts, miracidia and cercariae infection are dependent on different factors at both the individual and population level of their snail hosts.

Blood safety is a major element in the strategy to control the HIV epidemic. The aim of this study was to determine the prevalence and the associated factors of a positive HIV test among blood donors and its association between Human African Trypanosomiasis in Kikwit, the Democratic Republic of Congo. A cross-sectional study was conducted between November 2012 and May 2013. An anonymous questionnaire was designed to extract relevant data. The average mean age of participants was 30 years. The majority were man (67.8%). The overall prevalence of HIV, syphilis, hepatitis B, hepatitis C and human African trypanosomiasis was respectively 3.2%, 1.9%, 1.6%, 1.3% and 1.3%. Alcohol intake, casual unprotected sex, not using condoms during casual sex, sex after alcohol intake and seroprevalence of human African trypanosomiasis were significantly associated with a positive HIV test result ( p<0.05). In this study, sexual risk behaviors were the major risk factors associated with positive HIV tests in blood donors living in Kikwit. It is important to raise awareness about HIV and voluntary blood donation in response to some observations noted in this study such as the low educational level of the blood donors, the low level of knowledge of HIV prevention methods.

The aim of the present study was to determine the prevalence of antibodies to Toxoplasma gondii by two serological techniques in sera of 364 stray dogs from Brazil by immunofluorescence antibody test (IFAT, cut off point 1:16) and to the modified agglutination test (MAT, cut-off points 1:25 and 1:50). A total of 175/364 (48.07%) sera were positive by IFAT, and 108/364 (29.67%) and 85/364 (23.35%) were positive by MAT with cutoff points 1:25 and 1:50, respectively were positive by MAT. Cohen’s Kappa Coefficient between IFAT and MAT was 0.81 (excellent) and 0.66 (substantial) with cutoff points 1:25 and 1:50, respectively. Using IFAT as gold standard, MAT sensitivity and specificity were 78% and 99% for 1:25 and 61% and 99% for 1:50, respectively. The results document of the usefulness of MAT for serological diagnosis because it does not require species-specific conjugate.

There are few published studies on various vector borne diseases of dogs in India and most depict clinical infection in dogs, diagnosed by observation of the haemopathogens in stained blood smears. This study provides the first report regarding molecular confirmation and ancestral relationship analysis of blood smears positive cases of assorted haemopathogens in Punjab province of India. On blood smear examination, haemopathogens were observed in 124 out of 778 (15.95%, 95% CI: 13.53– 18.68) blood smears. Further polymerase chain reactions (PCR) was used on bloods smear positive cases to validate the results. Out of 778 blood samples, Babesia gibsoni was most common parasite infecting dogs (15.04%, 95% CI: 12.7–17.72), followed by Ehrlichia canis (0.39%, 95% CI: 0.0–1.13), infection of Babesia vogeli and Hepatozoon canis was same (0.26%, 95% CI: 0.0–0.9). Among various risk factors studied (age, sex, season), prevalence of infection was non-significantly higher in 1–2 year of age group (19.88%, 95% CI: 14.45–26.71), regarding sex same prevalence was recorded (15.94%), and chances of infection was highest in pre-monsoon i.e. summer (18.26%, 95% CI: 14.49–22.76). Phylogenetic analysis revealed ancestral background of Ludhiana isolates of B. vogeli , B. gibsoni, H. canis , and E. canis with the isolates of Philippines, Mongolia and Tunisia.

The present study describes a new species of Trichodina found in the mucus of the body surface of 137 farmed and wild silver catfish ( Rhamdia quelen ) located in southern Brazil and in the gills of 99 banded knife fish ( Gymnotus sp.) captured in the basin of Pantanal. Smears of the body surface mucus and gills were air dried at room temperature and impregnated with silver nitrate and stained with Giemsa for analysis of the adhesive disc, denticles and macronucleus. Characterized as a medium trichodinid, Trichodina quelenii n. sp. resembles T. heterodentata and T. acuta . It differs from T. heterodentata by the shape of the blade and from T. acuta by the lack of central circle. Furthermore, the distinct shape of the blade of T. quelenii n. sp. differs from all previously described species for the genus, leaving no doubt it is a new species.

Monocotyle luquei n. sp. (Monogenea: Monocotylidae) was described from gills of diamond stingray Dasyatis dipterura (Jordan and Gilbert, 1880) (Dasyatidae) off Peru. The new species can be differentiated from the other species of the genus by the combination of the following characteristics: (1) accessory sclerites on the dorsal posterior surface of the body absent, (2) only one testis is present, (3) 1–2 loops in the copulatory organ, (4) the male copulatory organ with a sclerotized accessory piece, (5) shape of five sclerites in marginal papillae, (6) size of anchor and (7) posterolateral septa bifurcated. This is the first record of species of Monocotyle Taschenberg, 1878 from the southern Pacific.

Nematodes, belonging to the family Thelastomatidae, are parasites of saprophytic terrestrial arthropods, mainly cockroaches. American cockroach Periplaneta americana (Linnaeus, 1758) has high environmental adaptability, and has been spread worldwide by human activity. There are several reports of thelastomatid parasitic nematodes of P americana in the world. Here, the thelastomatid nematode Hammerschmidtiella diesingi (Hammerschmidt, 1838) (Oxyuridomorpha: Thelastomatidae), isolated from P americana is recorded for the first time in Japan. Through morphometrics, DIC and SEM observations, we confirmed that specimens of the present study agree with previous records of H. diesingi . DNA sequencing of the partial D2/D3 LSU expansion segment of the 28S ribosomal RNA gene revealed that H. diesingi from Japan matches with H. diesingi from other countries. Our contributions are very useful and fundamental for further analysis of the cockroach and parasite relations.

Total 24 valid species belongs to 6 subgenera in genus Ixodes had been documented in China. Of them, Eschatocephalus (2 species), Scaphixodes (2 species); Partipalpiger (1 species), Pholeoixodes (3 species) Ixodiopsis (1 species) and the largest subgenus Ixodes (15 species) were involved. Fortunately, we found another 3 new record species, namely I. (Ixodes) apronophorus Schulze, 1924, I. (Ixodes)pavlovskyi Pomerantzav, 1946 and I. (Trichotoixodes) frontalis (Panzer, 1798), from the specimens deposited in medical entomology gallery of China, which increase a new subgenus Trichotoixodes to the tick fauna in China. In the present paper, the taxonomy characteristics of the new record species were described briefly based on the specimens examined and keys to the genus Ixodes were also updated.

The epizootic of Bluetongue virus (BTV) throughout Europe in 2006 revealed insufficient knowledge regarding seasonal activities, endo/exophilic and endo/exophagic behaviour of the species of the Culicoides genus (Diptera: Ceratopogonidae). Although several studies have been carried out in Western Europe, bringing new knowledge of the above mentioned topics, there was still a gap to fill in Central Europe. Therefore, this study investigated seasonal variations in the indoor/outdoor Culicoides activity observed in the south-eastern Slovakia from 2012 to 2014, using a light-trapping method. In total, 52,741 Culicoides were collected and identified; majority of them were caught outdoors (82.6%), with the highest activity period being mid-June. The C. obsoletus/C. scoticus and C.punctatus species predominated, altogether representing 88.8 and 94.1% of the total indoor and outdoor collections, respectively. Positive correlation was observed between the temperature and the abundance, whereas no correlation with other studied factors was detected. In autumn, Culicoides activity was observed outdoors at temperatures ranging between-1.5°C and 9.3°C, whereas the indoor activity was detected at temperatures ranging between-3.9°C and 0°C. This demonstrated that the most significant BTV vectors in the studied area are C. obsoletus/C. scoticus . The C. dewulfi and C. chiopterus vectors appeared to be of lower significance. The period with the highest risk of the BTV transmission in Eastern Slovakia appeared to be mid-June. The autumnal Culicoides activity inside the cowsheds may be important for the assessment of virus overwintering. This study is the first to provide the data on indoor/outdoor behaviour of biting midges in Slovakia.

Theileriosis caused by Theileria lestoquardi (malignant ovine theileriosis) in sheep and Theileria annulata (tropical theileriosis) in cattle is an important hemoprotozoal tick-borne disease in Iran. Due to major biologic and phylogenic similarities of these two species, this study was carried out to investigate the occurrence of natural infections with T.lestoquardi and T.annulata in cattle with clinical theileriosis in Ahvaz, southwest Iran. Fifty one cattle were selected based on clinical signs of theileriosis and confirmation by microscopic examination of blood smears. Blood samples were collected from each animal and hematologic and microscopic examinations were performed. Theileria piroplasmic forms were detected in all affected cattle. Pale mucous membranes (43.14%), icterus (11.76%) and fever (70.6%) were also observed. PCR-RFLP analysis revealed T. annulata infection in all tested cattle while coinfections with T. lestoquardi were found in two samples (3.92%). All sampled cattle including the two with mixed species Theileria infection were anemic. This is the first report of Theileria species cross infections in cattle with clinical theileriosis in Iran. It can be concluded that cattle can be infected with both pathogenic Theileria species, T. lestoquardi and T. annulata which can be an important issue in the epidemiology and spread of ovine malignant theileriosis.

Sera or meat juices of 177 red foxes ( Vulpes vulpes ) originated from the localities of a human-influenced landscape (Group 1) and 126 foxes from the protected mountain region (Group 2) of Slovakia, collected during 2010–2014 were tested for the presence of antibodies against Toxoplasma gondii using indirect ELISA and Neospora caninum by competitive ELISA. The tissue and uncoagulated blood samples were examined for the presence of the parasite’s DNA. The total seropositivity to T. gondii was 62.7% (190/303) and to N. caninum 26.4% (80/303). In the Group 1 antibodies to T. gondii were detected in 74.0% (131/177) and to N. caninum in 38.9% (69/177). In the Group 2 significantly lower seropositivity of 46.8% (59/126) to T. gondii antigens (P = 0.0218) and 8.7% (11/126) to N. caninum (P = 0.0001) was detected, respectively. However, by using molecular method, the presence of both parasites, was recorded less frequently. While in Group 1 T. gondii DNA was detected in 10.0% and N. caninum DNA in 18.3% of examined samples, in Group 2 T. gondii DNA was not detected at all and N. caninum was detected in 9.1% samples only. Results indicate that examined infections are highly common in the red foxes in Slovakia and are widespread in the locations of Eastern Slovakia bordering Poland, Ukraine and Hungary. The high infection rate in foxes representing reservoir hosts, presumably originates from their infected prey, ungulate carcasses, or from residual infected tissues in the hunting grounds after evisceration of shot animals during a hunting season.

Southern Italy, particularly Campania region, is an area where canine leishmaniasis (CanL) and zoonotic human visceral leishmaniasis (VL) are endemic. The red fox ( Vulpes vulpes ) has been hypothesized to play a role in occurrence of CanL in Italy but specific studies are poor. The aim of the present survey was to investigate the prevalence of Leishmania infection in dogs and foxes living in the same rural area (Picentini hills). 123 sera from autochthonous fox-hunting dogs were examined by immunofluorescent-antibody test (IFAT) using a cut-off of 1:160. The seroprevalence of dogs examined was 17.9%. Moreover, 48 foxes were examined after having been shooted by hunters or road accidents. Spleen, liver and lymph node samples were analyzed by specific Leishmania nested PCR (n-PCR). 10 foxes were found infected by L. infantum (20.8%) of which 4 animals in spleen, 2 in lymph nodes and 4 both in spleen and lymph nodes. The overall n-PCR positivity was 17.4% for spleen samples and 13.3% for lymph nodes; all liver samples resulted negative. In positive PCR foxes no signs clearly referable to leishmaniasis were recorded at necropsy. The results confirmed the presence of L. infantum infection in red foxes from Southern Italy, with a moderate level of exposure. Because large proportions of dogs with ascertained progressive leishmaniasis show a prolonged "subpatent condition" during which they are only positive to n-PCR before seroconversion, our results allow to assume that exposure risk in foxes is lower than hunting dogs living in the studied area.

Fascioliasis is a zoonosis actually considered as a foodborne trematode disease priority by the World Health Organization. Our study presents three cases of F. hepatica infection diagnosed by direct, indirect and/or imaging diagnostic techniques, showing the need of the combined use of them. In order to overcome some difficulties of the presently available methods we show for the first time the application of molecular tools to improve human fascioliasis diagnosis by employing a PCR protocol based on a repetitive element as target sequence. In conclusion, diagnosis of human fascioliasis has to be carried out by the combination of diagnostic techniques that allow the detection of infection in different disease phases, different epidemiological situations and known/new transmission patterns in the actual scenario.

Pathogenic strains of Acanthamoeba are causative agents of a sight threating infection of the cornea known as Acanthamoeba keratitis . AK cases have been reported in Iran recently due to inappropriate usage of contact lens maintenance and most patients report a contact with contaminated sources such as dust, water or soil. Sixty soil samples were collected from public and recreational areas in the province of East Azerbaijan, Iran and checked for the presence of Acanthamoeba spp. Samples were cultured on non-nutrient agar plates seeded with heat killed Escherichia coli . PCR and sequencing of the DF3 region were carried out in order to genotype the isolated strains of Acanthamoeba . Thermotolerance and osmotolerance assays were performed in order to investigate the pathogenic potential of isolated Acanthamoeba strains. Acanthamoeba spp. was isolated from 41.6% of soil samples and genotyping of the strains resulted in the identification of genotypes T3, T4, T5 and T11. Most of the isolates belonging to genotypes T3 and T4 showed high pathogenic potential, indicating that they might present a potential health hazard for humans and other animals in this region. To the best of our knowledge, this is the first report on the identification of genotypes T3 and T11 from soil sources in the country.

The veterinary important parasite of ruminants, giant liver fluke Fascioloides magna (Trematoda: Fasciolidae), isolated from liver of farmed fallow deer ( Dama dama ) from Podkarpackie Province (southeastern Poland) was genotypized by mitochondrial cytochrome c oxidase ( cox 1) and nicotinamide dehydrogenase ( nad 1) markers. The data on this newly emerged population were compared with mitochondrial haplotypes of recently detected Polish population of F. magna from Lower Silesian Wilderness (southwestern Poland) and with European populations of the parasite from all three natural foci; northern Italy, Czech Republic and the Danube floodplain forests. The flukes from Podkarpackie Province were found to be genetically identical with flukes from Czech Republic and Lower Silesian Wilderness in Poland. It is evident that central and southwestern Czech Republic, recognized as one of the endemic area of F. magna in Europe, has been enlarging and parasite has been invading several novel localities in Poland.

Free-living amoebae of Acanthamoeba genus are opportunistic pathogens distributed worldwide. Strains included in this genus are causative agents of a fatal encephalitis and a sight-threating keratitis in humans and other animals. In this study, 550 clinical samples which were collected between 1984 and 2014 from different patients with suspected infections due to Acanthamoeba were initially screened for the presence of this amoebic genus at the Laboratorio de Amibiasis-Escuela de Bioanálisis at the Universidad Central de Venezuela. Samples were cultured in 2% Non-Nutrient agar plates seeded with a layer of heat killed Escherichia coli . From the 550 clinical samples included in this study, 18 of them were positive for Acanthamoeba genus after culture identification. Moreover, positive samples were confirmed after amplification of the Diagnostic Fragment 3 (DF3) of the Acanthamoeba 18S rDNA genus and sequencing was carried out in order to genotype the isolated strains of Acanthamoeba . Furthermore, the pathogenic potential of the strains was checked by performing thermotolerance and osmotolerance assays. Sequencing of the DF3 region resulted in the identification of genotype T4 in all the isolated strains. Moreover, most isolates were thermotolerant or both thermotolerant and osmotolerant and thus were classified as potentially pathogenic strains. To the best of our knowledge, this is the first report on the molecular characterization at the genotype level of Acanthamoeba strains in Venezuela.

Parasites are ubiquitous members of biotic communities. To investigate the infective status of helminth parasites in Pelophylax nigromaculatus from Shanghai, 90 frogs were collected from July 2013 to July 2014 and examined for the presence of internal parasites. Of these, 86.67% (78/90) of frogs were infected with parasites, the total helminth intensity within infected frogs ranged from 1 to 367, and the mean intensity was 28.49. The infection rates and intensities were 78.89% and 22.89 for trematodes, 50.00% and 5.24 for nematodes, 13.33% and 3.92 for cestodes, and 41.11% and 8.49 for acanthocephalas, respectively. The majority (60.04%) of parasites were parasitic in the intestine, followed by urinary bladder (24.8%) and lungs (7.38%). Based on morphological features, 13 different species of helminth, including 9 undetermined species, were identified. The infective status of different species was significantly different. The most prevalent species were Diplodiscus nigromaculati (64.44%), Diplodiscus sp. (37.78%), Pomphorhynchus sp. (35.56%), Strongyloides sp. (33.33%). The mean infection intensity of Diplorchis nigromaculatus (139.25) was higher than the others’ species ranged from 3.57-14.63. This is the first reported discovery of Pomphorhynchus sp. (Pomphorhynchidae Yamaguti, 1939) in frogs from China. These data provide the foundation for further analyses of parasites in this and other species of amphibians.

The purpose of this study was to determine subjective ocular symptoms occurring in patients infested with Demodex . The number of Demodex mites in the obtained material that correlated with the appearance of ocular symptoms was estimated. The study material were eyelashes collected from 1499 patients. The material were observed under a light microscope. T-test, the logistic regression method, and Pearson correlation coefficient were used for the analysis. Demodex mites were detected in 47% patients. The mean ages of infected women and men were 64 and 59 years, respectively. 64% infected patients complained of one or more ophthalmological symptoms. The most commonly reported symptoms included itching (28%), redness of eyelids (21%), and watery eyes (15%). Positive correlation was found between itching, redness, pain, purulence or eyelash loss and the presence of Demodex . The mentioned symptoms increase the probability of Demodex infestation in a statistically significant manner (p<0.005). A correlation between the age and gender and the number of Demodex was revealed by the study. The threshold average number of seven Demodex mites per eight collected eyelashes with which the risk of the occurrence of an ocular symptom increases significantly was defined. In patients with a low number of Demodex mites, symptoms may be absent. The risk of the occurrence of ocular symptom in patients with demodicosis increases with the increase in the average number of Demodex mites.

Cystic hydatidosis is a re-emerging parasitic zoonosis with worldwide distribution. The current study was carried out to investigate the possible role of rats in the epidemiology of such disease in urban and suburban areas. For this purpose, a total of 50 feral Norway rats ( Rattus norvegicus ) were collected from urban and suburban settings, Cairo, Egypt. Rats were examined to be infected with cystic hydatidosis through serological examination by IHA test as well as post-mortem examination of internal organs, histopathological or molecular identification of the collected cysts. Moreover, 42 persons inhabiting suburban areas were tested for cystic hydatidosis by IHA. The overall seroprevalence rates of cystic hydatidosis in the examined rats and persons were 36% and 11.9% respectively. Cysts from 3 rats were identified as E. granulosus hydatid cysts (one via histopathological examination while the others by molecular technique and genotyped as G6 strain). The results of the current study highlight the possible role of Norway rat in the epidemiological cycle of E. granulosus especially in urban and suburban settings.

Numerous specimens of the parasitic nematode Rhabdochona papuanensis Moravec, Riha et Kuchta, 2008 (Spirurida: Rhabdochonidae) were collected from the intestines of the Australian endemic freshwater fish (eastern rainbow fish) Melanotaenia splendida (Peters) (Melanotaeniidae, Atheriniformes) in the Behana Creek, North Queesland during November of 2015. Although many species of Rhabdochona Railliet, 1916 are known to be common parasites of fishes in other continents, the present finding of R . papuanensis represents the first record of the species belonging to this genus from the Australian mainland. Light and scanning electron microscopical examinations of these newly collected specimens made it possible to redescribe in detail this nematode species, originally incompletely described from a congeneric host in Papua New Guinea. Fully developed, filamented eggs of R. papuanensis and the conspecific fourth-stage larva are described for the first time. The present finding of R . papuanensis in M . splendida from Australia represents new host and geographical records.

The pace of anti-malarial drug discovery is often impeded due to the lack of tools to determine the cidality of compounds in vitro . An anti-malarial compound must have a cidal mode of action, i.e. kill parasites, in order to quickly reduce parasite load. A static compound that merely inhibits growth must be identified early on in the discovery cascade. In this paper, we describe a high-throughput fluorescent assay for determination of the cidality of an anti-malarial compound. The assay works on the principle that cultures treated with a static compound will exhibit re-growth while treatment with a cidal compound leads to a marked reduction in parasite number. Parasite cultures are treated with the drug for 48 or 72 h following which the drug is washed off. Cultures are allowed to recover in drug-free media for 72 h and DNA content estimated using the fluorescent dye SyBR Green I. Following estimation of IC 50 and IC 99 values, we find that the IC 99 /IC 50 ratio is a reliable indicator of the cidality of a compound. Cidal compounds like artemisinin and chloroquine display an IC 99 /IC 50 ratio <5 while the ratio for a static compound like atovaquone is <5. This correlation holds true for various anti-malarial drugs with known modes of action. Importantly, the IC 99 /IC 50 ratio drops to <5 when a compound becomes cidal in action with longer duration of treatment. The assay is robust, reliable and provides a fast and effective means for prioritizing cidal compounds for progression along the drug discovery cascade.

During an extensive survey of fish parasites along the Malabar coast of India we observed exceptionally high or even massive infection of the narrow-barred Spanish mackerel, Scomberomorus commerson (Scombridae), with parasitic copepods Cybicola armatus . Such infection has not hitherto been reported. The copepods occurred on the gills of its host and the infection persisted for many years (the survey lasted from January 2010 to July 2014). The overall prevalence was very high (95%); out of a total of 981 fish studied, 933 were infected, and the maximum intensity reached 42. The highest rate of infection (prevalence 99.2%) was observed in medium sized fish (36.0-79.9 cm). We found this parasite only on S. commerson even though we examined 81 fish species, including five of the family Scombridae. Interestingly, the parasite exhibits strict site specificity and microhabitat preference. A significant proportion of them was seen attached to pseudobranchs rather than to main gill arches. Significantly, the body size of the parasite was positively correlated with that of the host fish. Our SEM micrographs contributed new facts to this copepod’s morphology by illustrating for the first time the ventral part of its cephalothorax, illustrating the nature of attachment, and by finding a new structure – a premaxillary pad. We demonstrated the haematophagous nature of C. armatus by finding and documenting the host’s blood cells in the parasite’s intestine.

The study was performed in the Trzciel Forest Districts which is located in the west part of Poland. The Scots pine is the main tree species creating forest landscape there. Dermacentor reticulatus , usually found in wet, boggy and damp habitat, in this case was discovered in fresh mixed coniferous forest and fresh coniferous forest. In Central Europe the Dermacentor reticulatus is after Ixodes ricinus the second most important vector for tick-borne diseases in Europe. The ticks were collected by flagging from lower vegetation during the autumn peak of their activity. All Dermacentor reticulatus were checked for presence of tickborne pathogens using PCR and nested PCR. In total 125 Dermacentor reticulatus ticks were collected. Among the pathogens examined, only Babesia microti has been found in 4% of them whereas Borrelia burgdorferi sensu lato and Anaplasma phagocytophilum have not been found. In this research the presence of Babesia. microti in Dermacentor reticulatus has been proved. It should be stressed that it is a new focus for the Dermacentor reticulatus tick in Western Poland.

Cutaneous leishmaniasis (CL) caused by Leishmania major is common in southern Israel, while Leishmania infantum (sub-strain of L. donovani , causing zoonotic visceral leishmaniasis) infections were rarely reported in Israel and only in other regions. We report the first case of L. infantum infection in southern Israel, presented atypically as CL in an immunosuppressed 47-year old male. The patient was treated with liposomal amphotericin-B and recovered, without extra-cutaneous complications. Diagnosis of L. infantum CL was confirmed by microscopic identification of amastigotes in Gimsa-stained smear of skin lesion, positive blood serology and a positive polymerase chain reaction (PCR) amplification of the internal transcribed spacer 1 genes (ITS1) and restriction fragment length polymorphism (ITS1 PCR-RFLP). We also review the medical literature on old-world CL caused by L. infantum . Multiple L. donovani/infantum CL cases were identified in the literature search. These can be divided schematically to two: 1) In several endemic countries, L. infantum strains are the main causative agents of CL; 2) In other regions, CL is almost exclusively caused by L. major or L. tropica , while L. donovani strains CL cases were reported sporadically or as imported disease.

A phenomenon of switching of the parasite in the food chain to an accidental host is commonly observed in nature. However, there is little available data concerning the morphological descriptions of parasites that passively get into the atypical hosts and are capable, at least to some degree, of somatic growth and development of reproductive structures. A morphological survey of Diplodiscus subclavatus (Pallas, 1760) adults isolated from a digestive tract of an accidental host, Viviparus contectus (Millet, 1813), was carried out. Diplodiscus subclavatus individuals identified in prosobranch snails were morphologically similar to adult forms of the parasite described from amphibians, typical final hosts in the life cycle of this paramphistomid. The observed forms of D. subclavatus had a fully developed reproductive system, sperm in the seminal vesicle and oocytes in the ovary. The number of eggs in the uterus ranged from 3 to 17. Our research indicates that D. subclavatus individuals reach the sexual maturity in the accidental, invertebrate hosts.
