Diabetic neuropathic pain, an important microvascular complication in diabetes, is recognised as one of the most difficult types of pain to treat. The development of tolerance, inadequate relief, and potential toxicity of classical antinociceptives warrant the investigation of the newer agents to relieve this pain. Reactive oxygen/nitrogen species, increased oxidative stress, cytokines, and apoptosis are implicated in the pathogenesis of diabetic neuropathy. The aim of the present study was to explore the effect of methanolic extract of aerial parts of H. spinosa (HSME) on alloxan induced diabetic neuropathy in Wistar rats. Diabetic rats developed neuropathy after the third week of diabetes induction. Chronic treatment with HSME (250, 500, and 750 mg/kg body weight; p.o.) for 6 weeks starting from the 3rd week of alloxan injection showed significant increase in the pain threshold levels as compared to diabetic rats. HSME treated diabetic animals showed significant decrease in blood glucose level and increase in body weight as compared to diabetic control animals. The changes in lipid peroxidation status and antioxidant enzymes levels observed in sciatic nerve of diabetic rats were significantly restored by HSME treatment. Thus, the results suggest therapeutic potential of H. spinosa in treatment of diabetic neuropathy.

Shea butter is traditionally used in Africa for its anti-inflammatory and analgesic effects. In this study we explored the anti-inflammatory activities of the methanolic extract of shea butter (SBE) using lipopolysaccharide (LPS)-induced murine macrophage cell line J774. It was observed that SBE significantly reduced the levels of LPS-induced nitric oxide, Tumor necrosis factor-α (TNF-α), interleukins, 1β (IL-1β), and -12 (IL-12) in the culture supernatants in a dose dependent manner. Expression of pro-inflammatory enzymes, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) were also inhibited by SBE. These anti-inflammatory effects were due to an inhibitory action of SBE on LPS-induced iNOS, COX-2, TNF-α, IL-1β, and IL-12 mRNA expressions. Moreover, SBE efficiently suppressed IκBα phosphorylation and NF-κB nuclear translocation induced by LPS. These findings explain the molecular bases of shea butter’s bioactivity against various inflammatory conditions and substantiate it as a latent source of novel therapeutic agents.

Infertility is the inability to conceive after one year of unprotected intercourse. In the present study, herbal composition prepared by using medicinal plants having aphrodisiac potentials was administered orally to the albino rats for 40 days and to the oligospermic patients for 90 days in order to prove the efficacy of herbal composition. Herbal composition was the mixture (powder form) of the medicinal plants namely, Mucuna pruriens (Linn), Chlorophytum borivillianum (Sant and Fernand), and Eulophia campestris (Wall). In the neem oil treated albino rats, there was significant reduction in almost all the parameters viz. body weight, testes and epididymes weight, sperm density and motility, serum levels of testosterone, FSH, and LH compared with control rats. Treatment with said herbal composition for 40 days results significant increased in the body weight, testis, and epididymes weight in rats. Concomitantly the sperm motility and the sperm density were significantly increased. After 90 days of treatment with this herbal composition, sperm density vis-a-vis motility was increased in oligozoospermic patients as a result of elevation in serum testosterone levels. No side effects were noticed during the entire duration of the trial.

The in vitro study of the antioxidant properties of the hydroalcoholic extracts of various Indian medicinal plants can logically help to develop a better and safer way of amelioration from oxidative stress. As aimed, the present study has been done to estimate and thereby conclude regarding the antioxidant activities of a few Indian medicinal plants, viz., Terminalia chebula, Terminalia belerica, Emblica officinalis, Caesalpinia crista, Cajanus cajan, and Tinospora cordifolia. The extracts of the plants have been subjected to the evaluation of antioxidant properties through scavenging assays for reactive oxygen species like superoxide, nitric oxide, peroxynitrite, hypochlorous acid, singlet oxygen, etc. and measurement of TEAC values and other phytochemical parameters. The phenolic and flavonoid contents of each plant have been found to be correlated to their individual antioxidant activity. The results showed the hydroalcoholic extracts of the plants were efficient indicators of their antioxidant capacity thus concreting their basis to be used as natural antioxidant.

This study was designed to examine the antioxidant defense by chloroform extract of Calotropis gigantea on streptozotocin-(40mg/kg, intraperitonial, single-injection) induced diabetes in wistar albino rats. The extract significantly (P < .05) decreased the pancreatic thiobarbituric acid-reactive substances (TBARS) levels and significantly (P < .05) increased the superoxide dismutase, catalase, and glutathione levels as compared to above levels in pancreatic tissue of pathogenic diabetic rats. The results of test drug were comparable to Glibenclamide (5mg/kg, daily), a standard antihyperglycemic agent. The study concludes that Calotropis gigantea enhances the antioxidant defense against reactive oxygen species produced under hyperglycemic condition and this protects β-cells against loss, and exhibit antidiabetic property.

Preliminary investigation of the in vitro and in vivo efficacies of different extracts from the leaves of Vernonia amygdalina (VA), a plant widely used in Nigeria was evaluated in Balb/C mice infected with a laboratory strain of Leishmania major (L. major). The ability of the methanol, hexane and aqueous extracts of the plant to suppress the infection rate and its cytotoxicity on macrophages and L929 cells were determined in the in vitro study. The in vivo study evaluated time course of infection, lesion progression and the histopathology of cutaneous lesions, liver and spleen after inoculation with metacyclic promastigotes. Methanolic extract of VA containing high levels of flavanoids, was the most potent extract as it showed the highest suppression on infectivity and viability of intracellular amastigotes at a concentration lower than that which elicited cytotoxicity on macrophages. Treatment of infected mice with methanolic extract of VA showed delayed onset of disease with a significant reduction in lesion size and attenuation of the histopathological outcome characterised by intact epidermis and less tissue destruction in skin, spleen and liver. In conclusion, these results demonstrate that VA has potent antileishmanial properties which warrants further investigation into the immunological mechanism.

The aim of this study was to investigate the antioxidant and anti-inflammatory potentials of crude extracts of Lopholaena coriifolia (Sond.) E. Phillips & C.A. Sm. and its isolated compounds. Separation and structure elucidation of Lopholaena coriifolia (Sond.) E. Phillips & C.A. Sm. were conducted using chromatographic and spectroscopic method. The antioxidant activities of the extracts in this study were determined by the ferric reducing antioxidant power (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and β-carotene bleaching assays meanwhile the anti-inflammatory activity was evaluated using the 5-lipoxygenase assay. Seven known compounds quercetin 3-O-glucoside (1), naringenin 7-O-glucoside (2), seneciphylline-O-glucoside (3), chrysoeriol (4), retrorsine (5), adonifiline (6) and 5,4′-di-O-methyl alpinumisoflavone (7) were isolated from ethanol extract of Lopholaena coriifolia (Sond.) E. Phillips & C.A. Sm. The ethanol and water extracts of Lopholaena coriifolia (Sond.) E. Phillips & C.A. Sm. elicited potent antioxidant and anti-inflammatory properties. Amongst the isolated compounds quercetin 3-O-glucoside gave strong antioxidant activity and adonifiline strongly inhibited 5-lipoxygenase activity.

Bio-efficacy of n-hexane fraction of sepal of Salmalia malabarica was evaluated covering the biochemical sensors for the management of hyperglycemic and hyperlipidemic effects. Evaluation of n-hexane fraction of Salmalia malabarica (SMH) from hydro-methanolic (2:3) extract at the dose of 0.1 gm/kg body weight twice a day were investigated in normal and streptozotocin (STZ) induced diabetic rats. Normal and STZ-induced diabetic rats were divided into five groups. The effect of the fraction on fasting blood glucose (FBG), serum insulin, hemoglobin, glycated hemoglobin, total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDLc), low density lipoprotein cholesterol (LDLc), very low density lipoprotein cholesterol (VLDLc), phospholipids, free fatty acids, urea, uric acid, creatinine, albumin and transaminases were investigated in STZ-induced diabetic rat. A significant reduction of FBG level was observed after SMH treatment in STZ-induced diabetic rat. Treatment of diabetic rats with n-hexane fraction of this plant restored the levels of the above biochemical sensors significantly (p<0.001) in respect to the control. Histological studies of pancreas showed a qualitative diminution in the area of the islet’s of Langerhans in diabetic group which was recovered by said fraction. Phytochemical screening of the fraction revealed the presence of flavonoids, terpenoids and steroids.

Although some herbal remedies in association with vitamin therapy have been investigated in eradicating HP, no research has been done to investigate the effects of lycopene it. Our aim was to understand if lycopene could be effective in eradication of HP. In this parallel group quasi-control trial, a total of 54 patients whose diagnosis of HP had been confirmed by rapid urease test (RUT) were enrolled. Group 1 received the standard 4-drug therapy to eradicate HP (Metronidazole 500 mg/BD, Amoxicillin 1g/BD, Omeprazole 20mg/BD, and Bismuth 240 mg/BD) and group 2 received the same regimen in association with Lycopene (30 mg/daily). One month after the initiation of the treatment, the patients were evaluated for HP eradication by RUT. Although eradication rate was higher in the second group, bivariate analysis showed no significant statistical difference between the two groups. In contrast with other nutrients, it seems that Lycopene does not have any significant effects on eradicating HP in comparison with the standard antibiotic therapy. The prevalence of HP is in association with socioeconomic situation, so the patients in different studies should be paid more attention about their own life style. We recommend that more studies can be designed by considering control group and placebo administration.

Typhoid is one of the most serious infectious bacterial diseases in third world countries. It is usually treated by traditional antibiotics but due to the appearance of antibiotic resistant strains physicians opt for phyto products and other alternative medicines for the treatment of typhoid. Baptisia, an extract from indigo plant root, has been proved to be highly effective ultradilute medicine for the treatment of typhoid; however, the mode of action of the ultradilute extract is uncertain. Due to the antigenic variations of Salmonella it seems to induce immuno system by activating both T and B cells by the formation of antibodies. This principle seems to be highly effective for the development of typhoid vaccine. The present studies found that Baptisia administration possibly caused a salmonella-like reaction in the body as this extract produces an endogenous antibody similar to salmonella reaction. Thus, this study suggests that Baptisia tinctoria extract can be used for the prevention and treatment of typhoid.

The aim of this study was to determine the possible underlying mechanism of the hypoglycaemic activity of the ethanolic extract of Nauclea latifolia leaves in rats. The extract, glibenclamide or water was administered orally in a glucose, sucrose and maltose tolerance tests. In addition, the effect of the extract on α-glucosidase enzymes was also studied. The ethanolic extract at 200mg/kg body weight inhibited the increase in glucose level after both oral and intraperitoneal glucose loads as did glibenclamide. The extract also dose dependently inhibited both maltase and sucrase activities in vitro but not in vivo. The hypoglycaemic effect of N. latifolia leaf ethanolic extract thus appears to be most probably exerted through a mechanism similar to that of glibenclamide which is related to increased insulin release from pancreatic β-cells.

Spirulina platensis (Spi) is a microalga presenting high contents of proteins, γ-linolenic acid, vitamins and minerals, and showing many biological activities. It is a promising drug for the treatment of diseases including diabetes. The objectives of this work were to study Spi effects on alloxan-induced diabetic rats, and associate this to its anti-inflammatory activity. The treatment with Spi (25, 50 or 100 mg/kg, p.o.) started 48 h after the alloxan injection, continuing for 5 or 10 days. Biochemical parameters were measured in sera of treated and untreated animals. The anti-inflammatory activity of Spi was assessed by the formalin test and carrageenan-induced paw edema in mice. Immunostainings for TNF-alpha were carried out in the carrageenan-induced paw edema in rats, before and after the Spi treatment, and its effect on the release of myeloperoxidase from human neutrophils was also determined. Spi decreased glycemia as well as triglyceride and total cholesterol levels of diabetic rats. Levels of urea and creatinine were also reduced, while liver transaminases were unaltered. Spi also decreased dose-dependently the 1st (neurogenic) and mainly the 2nd phase (inflammatory) of the formalin test, as well as the carrageenan-induced paw edema in mice. The anti-inflammatory effect of Spi was further confirmed by decreases in TNF-alpha immunostaining in the inflamed paw and in the myeloperoxidase release from human neutrophils. The results showed that the anti-diabetic effect of S. platensis is already manifested after a 5-day treatment. Additionally, considering the relationship between diabetes and inflammation, the microalga anti-inflammatory action may also be involved.

The study was aimed to investigate antiulcer effects of acetone extract and its different fractions Mimusops hexandra against experimental gastro-duodenal ulcers. 80% acetone extract of stem bark of Mimusops hexandra (Extract A, p.o.) and its different fractions namely diethyl ether (Extract A1, p.o.), ethyl acetate (Extract A2, p.o.) and aqueous (Extract A3, p.o.) were tested for the presence of preliminary phytoconstituents and were screened for their antiulcer potential in experimental animals using ethanol-HCl and aspirin-induced gastric damage at the dose of 500 mg kg–1p.o. Extract A2 being the most active fraction amongst all the fractions tested was also studied at different doses to find its ED50. Further, to establish the mechanism of action, Extract A2 was also tested for its effects in acetic acid-induced gastric ulcer models and cysteamine-induced duodenal ulcer. The effect was compared with cimetidine. Flavonoids (quercetin), procyanidins, saponins and triterpenoids were found to be present in bark. Oral administration of Extract A2 inhibited formation of gastric lesions induced by aspirin in a dose dependent manner. Elevated level of lipid peroxidation due to ethanol-HCl and aspirin induced gastric damage was significantly (p<0.05) reduced by the treatment with Extract A2. Further, Extract A2 at the dose of 100 mg kg-1 (p.o.) reduced extent of acetic acid induced gastric ulcer in experimental animals. Moreover, protection afforded by Extract A2 against cysteamine-induced duodenal lesions was evident from dose dependent decrease in ulcer index (p<0.05), score for intensity (p<0.05) and total lesion area (p<0.05), when compared with the control group. The antiulcer activity shown by Extract A2 in experimental gastro-duodenal ulcer could be attributed to decrease in gastric acid secretory activity along with strengthening of mucosal defensive mechanisms.

Introduction: Blood platelets are directly involved in both haemostatic and pathologic thrombotic processes, through their adhesion, secretion and aggregation. In this study, we investigated the effect of genins (aglycone flavonoids without sugar group) isolated from parsley (Petroselinum crispum) leaves in vitro on human platelet aggregation and adhesion to a collagen-coated surface under physiologic flow conditions. Materials and Methods: The aggregation and adhesion studies were monitored after pre-incubation of platelets with genins. Results: Genins inhibited dose dependently aggregation induced by thrombin, ADP and collagen. The strongest effect was observed in collagen induced aggregation (IC50 = 0.08 ± 0.01 mg/ml). The HPLC identification of genins compounds revealed the presence of keampferol, apigenin and other not identified compounds. The aggregation tests showed that these compounds have anti-aggregating activity. In addition, adhesion of human platelets to collagen was greatly decreased (over 75 %) by genins (0.3 mg/ml). While the mechanism by which genins act is unclear, we suggest that these compounds may interfere with a multiple target step in the haemostasis process. Conclusion: These results show that genins isolated from parsley has a potent antiplatelet activity. It may be an important source of beneficial antiplatelet compounds that decrease thrombosis and cardiovascular diseases.

We have investigated on the potentiation of etoposide (ETP) and temozolomide (TMZ) cytotoxicity in U-87MG glioblastoma and D283 medulloblastoma cell lines by curcumin (CUR) and turmeric force (TF), a nutraceutical formulation of turmeric, with the objective of assessing the potential for their adjuvant use in brain tumor chemotherapy. While U-87MG cell line was generally resistant to TMZ, IC50 values for CUR and TF were 37.33 and 30.75 µg/ml, respectively. TF is the only agent that demonstrated efficacy at the IC90 level. When CUR or TF was combined with ETP and TMZ, increased chemotherapeutic efficiency in the U-87MG cells was observed. TF is highly cytotoxic to D283 Med cell line compared to curcumin with an IC50 value of 1.55 ug/ml. Although both CUR and TF potentiated ETP and TMZ cytotoxicity, TF is more efficient than CUR in both U-87MG and D283 Med cell lines. Treatment of U-87MG cells with the triple combination of TMZ+ETP+TF induced a high percentage of apoptotic cells. Potential mechanisms that may explain evidence of synergy include down regulation of p10 and p53 mRNAs and increase in BAX/Bcl-2 mRNA ratio. These pre-clinical results suggest that TF may be useful as an adjuvant with ETP and TMZ for brain tumor chemotherapy.

Exposure of rampant higher glucose level in diabetic condition could lead earlier and late diabetic complication eventually caused malign effect in body organs. The aim of the present work was to analyze the anti-diabetic potential of chloroform fraction of ethanol extract of Andrographis paniculata (AP) and diabetes laden gene expression alteration. In streptozotocin (60 mg/kg bw, i.p.) induced Type 2 diabetes albino mice, two weeks consecutive repeated dose treatment of chloroform fraction of AP at dose of 200 mg/kg, orally was evaluated for antidiabetic screening. Fasting blood glucose, oral glucose tolerance, serum lipid profile, tissues glycogen content, glucose 6-phosphatase and hexokinase enzymes level in liver, in vitro and in vivo insulin estimation were measured on last day of treatment. Subsequently presence of responsible phytoconstituents for respective activity was observed by HPLC. In diabetic mice, the fraction reduced the level of harmful cholesterol, while increased the favorable cholesterol significantly. Biochemical enzymes like glycogen, glucose-6-phosphatase and hexokinase were evaluated in body tissues. Apart from this in vitro, in vivo insulin estimation and diabetogenic gene expression analysis like GK, PEPCK, G-6pase, Glut-4, AR, PPAR-α,γ and TNF-α were evaluated using RT-PCR technique. Antidiabetic screening of fraction of AP at molecular level revealed significant antidiabetic activity.

Cucumis melo Linn. (Cucurbitaceae) fruits have been used, traditionally in Indian traditional system of medicine, for the treatment of various disorders such as liver tonic, cardioprotective, antidiabetic, antiobesity, etc. The aim of the present study was to investigate the possible anti-hyperlipidemic activity of Cucumis melo fruit peel (CMFP) methanolic and aqueous extract in high cholesterol diet induced hyperlipidemia in rats. Treatment with CMFP methanolic and aqueous extract showed significant (P<0.01) reduction in gain in body weight, serum lipid profile like total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) level, atherogenic index and increased the serum high density lipoprotein cholesterol (HDL-C) levels in 28 days treatment when compared to the hyperlipidemic control group. The fecal excretion of bile acids and sterols was further increased upon treatment with CMFP methanolic and aqueous extract and standard drug. Administration of methanolic extract of CMFP at a dose of 500 mg/kg showed higher antihyperlipidemic activity as compared to other extract treated groups. The results concluded that CMFP methanolic extract (500 mg/kg) have potent antihyperlipidemic activity in high cholesterol diet induced hyperlipidemia model and which is equipotent activity when compared with atorvastatin treated group.

Objective: The present investigation was aimed at discovery of novel acetylcholinesterase (AChE) inhibitors.Methods: In vitro AchE inhibitory activity of various extracts of Sphaeranthus indicus flower heads was carried out. The petroleum ether fraction of S. indicus flowers (SIPE) exhibited significant activity. The fraction was found to be rich in sesquiterpene lactone content possibly responsible for in vitro AChE inhibition. In further study, the antiamnesic activities of SIPE in mice on the learning and memory impairments induced by scopolamine (1.0 mg/kg, i.p.) were examined.Results: SIPE (10 mg/kg, p.o.) administration significantly reversed cognitive impairments in mice by passive avoidance test (P < 0.05). It also reduced escape latencies in training trials and prolonged swimming times in the target quadrant during the probe trial in the water maze task (P < 0.05). These results indicated that S. indicus due to its sesquiterpene lactones have anti-cholinesterase activity. A major sesquiterpene lactone, 7-hydroxy frullanolide along with other constituents were isolated from SIPE and evaluated for AchE inhibitory activity. Negative results were obtained in case of isolated compounds.Conclusion: Synergistic effect between constituents of SIPE was confirmed to have anti-amnesic activities that may be useful for cognitive impairment treatment.

Potentilla fulgens has been used for a long time as folk remedy for a variety of ailments without having information on its pharmacological action. The study was intended to determine the effectiveness in antitumor activity among kaempferol (KMP) as flavonoids, ellagic acid (ELA) as polyphenols and methanolic extract of the root of P. fulgens (PRE) in Ehrlich ascites tumour (EA) and MCF-7 cancer cells. The total phenolic and flavonoid content were found to be 138.8 ± 1.6 mg gallic acid and 401.6 ± 4.6 mg quercetin per 1 gm of the extract, respectively. The extract resulted in increasing in vivo survivality of mice bearing EA cells and loss of cell viability in a dose-dependent manner in MCF-7 cells. This effect may be attributed to apoptotic cell death as confirmed by flow cytometric analysis and PARP1 proteolysis. Such induction of apoptosis was achieved by suppression of inhibitor of the apoptosis protein survivin. It was observed that endogenous level of glutathione was depleted significantly in MCF-7 cells after the treatment with PRE only but not with KMP or ELA. This induction of apoptotic cell death and lowering the level of glutathione are highly desirable mode for an anticancer agent.

The bioactive properties of Manuka honey are now well recognised, but the nature of honey (a sticky, viscous fluid) can make it hard to use as a health remedy. A new technology using encapsulation of Manuka honey with alpha-cyclodextrin molecules has been developed, creating a free-flowing powder that can easily be added to foods and beverages, or tableted / made into capsules for use in health. In this study, we investigated for antioxidant and anti-inflammatory activity. Antioxidant capacity of raw Manuka honeys and matched complexes was measured using the CUPRAC method. Results showed that the antioxidant activity of honey decreased when complexed, this being directly related to dilution of the final product with alpha-cyclodextrin. Anti-inflammatory activity was assessed by measuring inhibition of neutrophil TNF-alpha secretion. Contradictory results were produced, with both stimulation and inhibition of TNF-alpha being observed. Data from this study indicate that the formation of cyclodextrin-based complexes of Manuka honey may potentiate the anti-inflammatory activity of honey, but this may differ depending on methylglyoxal content and the presence of other factors.

The present study investigated the effects of Gmelina arborea hexane leaves extract on markers of oxidative stress and its vasorelaxant effects on isolated rat aorta, in order to postulate the possible mechanisms involved in the antihypertensive properties of the plant. To evaluate the antioxidant effects of the extract, rats were randomly divided into four groups of five rats each. With the exception to the group receiving Tween (2.5%), the other groups were treated either with NaCl (900mg/kg/day) alone, NaCl (900mg/kg/day) combined with vitamin C (5mg/kg/day) or Gmelina arborea extract (150mg/kg/day). At the end of eight weeks of treatment, animals were sacrificed and some organs as well as blood samples were collected for biochemical analysis. The in vitro vasodilating effects of the extract (0.5-1.5mg/ml) were evaluated using intact and denuded rat thoracic aortic rings or aorta pre-incubated in L-NAME (2µM), indomethacin (2µM) or glibenclamide (2µM) and contracted with phenylephrine (1µM). The in vivo effects of G. arborea hexane extract prevented both left ventricular and vascular hypertrophy, it also modulated lipid metabolism. Moreover, the extract prevented lipid peroxidation, increased superoxide dismutase and catalase activity as well as NO level. On isolated rat aortic rings, the extract induced concentration-dependent vasorelaxant effects. Extract-induced vasodilation was reduced by mechanical denudation of the endothelium as well as pre-treatment with L-NAME, indomethacin or glibenclamide. These results indicate that Gmelina arborea hexane extract possesses bioactive compounds with antioxidant and vasorelaxant properties.

PURPOSE: To determine the effects of 6wk of supplementation with fish oil (FO) on blood pressure and the morning salivary cortisol concentration in normotensive adults. METHODS: Testing was performed following an overnight fast. Subjects (n=40; 35+/-13y, mean+/-SD) rested supine for 40min, at which time blood pressure and heart rate were measured. Saliva was collected and analyzed for cortisol. Subjects were then randomly assigned to either: 4g/d of Safflower Oil (SO); or 4g/d of FO supplying 1,600mg/d eicosapentaenoic acid and 800mg/d docosahexaenoic acid. Testing was repeated following 6wk of treatment. RESULTS: Compared to SO, there was a significant decrease in systolic blood pressure with FO (SO=1.3+/-5.8 mmHg; FO=-6.8+/-10.2 mmHg; p=0.004), a significant reduction in pulse pressure (SO=0.2+/-7.8 mmHg; FO=-6.4+/-8.8 mmHg; p=0.02), and a tendency for a decrease in mean arterial pressure (SO=1.2+/-5.3 mmHg; FO=-2.5+/-7.3 mmHg; p=0.08). There was a tendency for salivary cortisol to decrease with FO (SO=0.005+/-0.129 µg/dL; FO=-0.068+/-0.148 µg/dL; p=0.072), however, this change was not significantly correlated with the change in systolic blood pressure (r=0.021, p=0.929). CONCLUSION: 6wk of supplementation with FO significantly decreases systolic blood pressure in normotensive adults and this change was not significantly correlated with a reduction in salivary cortisol.

Context: Smilax zeylanica L.(Smilacaceae) is a climbing shrub with woody prickly stems. Objective: This study evaluated the hepatoprotective effect of S. zeylanica against paracetamol induced hepatotoxicity in Wistar rats. Materials and methods: The protective effects of the methanol extract (200 and 400 mg/kg) of root and rhizome of S. zeylanica were studied on paracetamol induced (1 g/kg) hepatic damage in Wistar rats by estimating the serum levels of AST, ALT, alkaline phosphatase (ALP), total proteins, total bilirubin and albumin. Sections of liver were observed for histopathological changes in liver architecture. Results: Rats were protected from the hepatotoxic action of paracetamol as evidenced by the significant reduction in the elevated serum levels of ALT (P<0.001), AST (P< 0.01, P< 0.001), ALP (P<0.05, P< 0.001), total bilirubin (P< 0.05) and an increased level of total protein (P< 0.05) and albumin (P< 0.05, P<0.01) with a significant reduction in liver weight (P<0.001), when compared with the paracetamol control. Silymarin (100 mg/kg) was used as the standard. Discussion and conclusion: The biochemical observations were supplemented by the histopathological studies on the liver sections of different groups. The methanol extract of S. zeylanica was found to alter the damage caused to hepatocytes by paracetamol and prevent the leakage of vital serum markers, which confirmed the hepatoprotective effect of this plant.

The methanolic root bark extract of Afzelia africana was tested for antidiabetic activities in-vivo. The acute toxicity of the extract was tested in mice and the result showed that the extract has low toxicity. Investigation on the phytochemical constituents of the plant extract revealed the presence of flavonoids, tannins, alkaloids, steroids and saponins. The plant extract was tested for antidiabetic activities in alloxan - induced diabetic mice at doses of 62.5 mg/kg, 125 mg/kg, 250 mg/kg, 500 mg/kg and 1000 mg/kg over a period of 6 hours. The activity was found to be both dose and time dependent. Optimum activity was noted at the dose of 250 mg/kg and 6 hours post treatment. Distilled water (10 ml/kg) and glibenclamide (2 mg/kg) were used as negative and positive controls respectively. The extract showed antidiabetic activity which did not differ significantly (p > 0.05) from glibenclamide.Column and thin layer chromatography revealed the presence of 5 fractions in the plant extract out of which fraction 3 was found to be active.

This study aimed to investigate the effects of a health product Squina (Diacylglyceryl Ether) (DAGE) on innate and adaptive immunity in mice. Both ex vivo/ in vivo mouse models and an in vitro system using cultured mouse splenocytes were recruited for the assessment of innate and adaptive immunity. For the innate immune response, DAGE extract treatment enhanced the LPS-induced IL1-β secretion by activated macrophages in vitro and long-term DAGE treatment (0.18 and 1.8 g/kg × 20 doses) elevated the phagocytic activity of macrophages, as well as natural killer cell activity in mice. The DAGE-induced increase in macrophage phagocytic and natural killer cell activities were accompanied by the suppression of tumor growth in Sarcoma-180 cell-inoculated mice. For the adaptive immune response, long-term DAGE treatment enhanced the splenocyte index and concanavalin A- stimulated proliferation ex vivo in mice. Consistently, the incubation with DAGE extract potentiated the concanavalin A-stimulated proliferation in mouse splenocytes in vitro. In conclusion, the results show that long-term DAGE treatment produces stimulatory effects on both innate and adaptive immunity in mice.

Glucosinolates (thioglucoside-N-hydroxysulphates) constitute a homogeneous class of naturally occurring thiosaccharidic compounds mainly found in the botanical order Brassicales. They can be hydrolyzed by myrosinase to produce D-glucose and various other degradation products like isothiocyanates (ITCs)-depending on the aglycon part. The exact function of glucosinolates (GLSs) in the plant is unclear, however their potent odour and taste suggests a role in herbivore and microbial defense. They are known for their fungicidal, bacteriocidal, nematocidal and allelopathic properties and have recently attracted intense research interest because of their cancer chemo-protective attributes. Iso-thiocyanates, one of the hydrolyzed products, show best anti-carcinogenic activity.

The health workforce is perceived to be in short supply in most developed and developing countries. There are concerns that this could result in reduced coverage of health services and the delivery of suboptimal care. Strategies to address the health workforce shortage have focussed predominantly on recruitment and training, with relatively little regard to the equally important issue of retention. One approach that may improve job satisfaction, opportunities for specialisation and the workload of health workers, and thus, improve retention and more importantly, patient outcomes, is role substitution. Many complementary and alternative medicine (CAM) practitioners appear to be well placed in terms of educational preparation to substitute (either horizontally or vertically) a number of roles traditionally performed by conventional health disciplines. As well as the potential benefits to the health workforce and the quality of patient care, role substitution could provide an important first step toward integrating CAM practitioners into mainstream health care settings.

Raphanus sativus (abbreviated in this paper as Rs.Cr) and Areca catechu (Ac.Cr), commonly known as radish and betel nut respectively, are traditionally used in South Asia for different gastrointestinal, gallbladder, and hepatic diseases. There has not been any study to see how they modulate gallbladder contractility. We selected isolated rabbit gallbladder tissue preparations, mounted them in tissue baths containing Krebs-Henseleit solution at 37 °C, and then recorded the changes in baseline tone of the tissues upon administration of Rs.Cr and Ac.Cr. Both the extracts exhibited concentration-dependent stimulant effect on the baseline tone of gallbladder tissues, similar to carbachol, a muscarinic receptor agonist. The stimulant effect of the extract, as well as that of carbachol, was completely blocked in the presence of atropine, a muscarinic antagonist, indicating similarity in the mechanism of action of the extracts with carbachol. The result shows potential of these extracts to contract the gallbladder and to subsequently increase bile secretion, but this remains to be investigated in detail. This study justifies the traditional use of radish and betel nut in different gastrointestinal disorders.

Objectives: To determine regular use of alternative remedies amongst undergraduate students. Design: Questionnaire covered use of alternative remedies as well as demographic details. Settings/Location: Online completion of the questionnaire. Subjects: Sixty-nine participants (55 female, 14 male) aged between 18 to 30 years studying undergraduate psychology at Bournemouth University, Dorset, UK. Interventions: Structured open and closed questions; all questions were given to all of the participants. Outcome Measures: Qualitative comments were captured as well as percentage of respondents according to answers collected. Results: Most respondents had used remedies for exam "anxiety" and obtained these products from relatives or friends or relatives. Conclusions: Reasons of effect provided by participants were that they provided a genuine effect; a general effect; or a placebo effect.

The antiulcer activity of 70% methanolic extract of fruits of Terminalia belerica Roxb. (Combretaceae) was evaluated using ethanol induced, aspirin induced, cold stress restraint and pylorus ligated ulcer in rats. The methanolic extract (100, 250, 500, 1000 mg/kg, p.o.) significantly suppressed the peptic ulcer induced by ethanol. Methanolic extract at a dose of 500 mg/kg showed, promising antiulcer activity hence, this dose was selected for further evaluation of antiulcer studies. The methanolic extract (500 mg/kg) showed significant (p<0.05) reduction in gastric volume, free acidity, total acidity, ulcer index, protein and pepsin content and increase in mucus content in pylorus ligated rats as compared to control. Treatment with Terminalia belerica methanolic extract further provided significant antiulcer protection against aspirin induced ulcers but not in cold stress restraint model. These results suggested that the 70% methanolic extract of Terminalia belerica increased resistance to necrotizing agents, providing a direct protective effect on the gastric mucosa and exhibited antiulcer effect.
