Many plants are found to possess reliable pharmacological properties and have started to attract the attention of researchers. One such holistic plant is Acorus calamus , commonly known as sweet flag, belonging to the rhizomatous family Acoraceae. The different parts of this plant, such as the leaves and rhizomes, are used traditionally in different medicinal preparations for the treatment of various ailments including arthritis, neuralgia, diarrhoea, dyspepsia, kidney and liver troubles, eczema, sinusitis, asthma, fevers, bronchitis, hair loss, and other disorders. Many reports have also appeared in mainstream scientific journals confirming its nutritional and medicinal properties. Biochemical analysis of the plant has revealed a large number of secondary metabolites that may be responsible for its rich medicinal properties. Basic scientific research has uncovered the mechanisms by which itexerts its therapeutic effects. Medicinal herbs such as A. calamus are quite promising in the recent therapeutic scenario, with a large number of people favouring remedies and health approaches that are free from the side effects often associated with synthetic chemicals. In this review, we try to summarise the ethno-medicinal uses, botanical descriptions, phytochemical constituents, and biological activity of the plant parts, as well as the molecular targets of A. calamus , which we hope will serve as a good base for further work on this plant.

Background: Menopause-associated estrogen deficiency results in climacteric symptoms like vasomotor, psychological and sleep disturbances that cause a decline in the quality of life. Electrodermal activity (EDA), a psychophysiological measure, reflects sympathetic activity, which provides information associated with individual’s emotions, phobias, arousal, cognition and stress. The study compared electrodermal response to auditory stimuli between postmenopausal and perimenopausal women with and without symptoms and also correlated the association of scores of the menopausal transition symptoms with indices of EDA. Methods: Seventy-five women volunteers in the age group of 45–60 years, 25 in each group who were postmenopausal, perimenopausal with symptoms and perimenopausal without symptoms, were recruited. Indices of EDA such as latency, amplitude, rise time and half recovery time for auditory stimuli were quantified using standard techniques. Symptoms of menopausal transition were assessed using Women Health Questionnaire. Results: Analysis using one-way analysis of covariance after controlling for variables showed that mean skin conductance level of EDA was significantly higher among perimenopausal women with symptoms compared with perimenopausal women without symptoms and postmenopausal women. Perimenopausal women with symptoms had significantly lower latency of response when compared with other groups. Analysis using Pearson correlation test showed that latency of EDA had significant positive correlation and amplitude had significant negative correlation with menopausal transition symptom scores. Conclusions: Perimenopausal women with symptoms exhibited increased sympathetic sudomotor activity when compared with perimenopausal women without symptoms and postmenopausal women as measured by EDA. Further, select measures of EDA exhibited significant association with the symptoms of menopausal transition.

Background: The first-line antituberculosis (anti-TB) drugs, isoniazid (INH), rifampicin (RIF), ethambutol (EMB), and pyrazinamide (PZA), are effective in the treatment of pulmonary tuberculosis. However, the toxicity of these drugs in the clinical setting limits their use. Here, we evaluated the effects of anti-TB drugs on the reproductive system in female rats. Methods: Thirty-five female Wistar rats were assigned into five groups of seven animals each. The control group received normal saline, whereas others received INH (5 mg/kg), RIF (10 mg/kg), EMB (15 mg/kg), and PZA (15 mg/kg) through gavage thrice a week for 8 consecutive weeks. Results: Administration of anti-TB drugs significantly (p<0.05) reduced uterine and ovarian weight, as well as the relative weight of the uterus when compared with controls. In addition, anti-TB drugs increased the activities of alanine aminotransferase as well as the level of total bilirubin. Treatment with INH, RIF, and PZA significantly (p<0.05) reduced the levels of follicle-stimulating and luteinizing hormones, estrogen, and prolactin. The INH, RIF, EMB, and PZA caused significant (p<0.05) increases in uterine malondialdehyde (MDA) levels by 281%, 214%, 273% and 190%, respectively, whereas INH and EMB increased the ovarian malondialdehyde by 111% and 129%, respectively. These drugs significantly (p<0.05) decreased the activities of ovarian glutathione-S-transferase and uterine glutathione peroxidase, superoxide dismutase, and catalase. Histology revealed the erosion of uterine mucosa, debris in the lumen of the uterus, congestion, and underdeveloped follicles in ovaries. Conclusions: The first-line anti-TB drugs elicited reproductive toxicity in the uterus and ovaries of rats through mechanisms that involved oxidative stress.

Background: Humans are directly/indirectly exposed to hazardous chemicals from the aquatic environment. We investigated the protection of the Launea taraxacifolia methanolic extract (LTME) on the hydroxyl steroid dehydrogenases [(∆ 5 -3β-hydroxyl steroid dehydrogenase (∆ 5 -3β-HSD) and the ∆ 5 -17β-hydroxyl steroid dehydrogenase (∆ 5 -17β-HSD), testicular 5′-nucleotidase and lactate dehydrogenase (LDH)] activities as well as the key indicators of oxidative stress in germinal epithelial cells of rats induced with surulere polluted river water (SPRW). Methods: The animals were divided into six groups (n=8). Group I was given 1 mL of distilled water only, Group II received 1 mL of SPRW only, Group III received 200 mg/kg LTME before+1 mL of SPRW after, Group IV received 200 mg/kg LTME+1 mL of SPRW, Group V received 1 mL of SPRW before+200 mg/kg LTME after and Group VI received 200 mg/kg LTME only. The treatment was done via oral administration for 28 days. Results: The HPLC results showed the abundance of quercetin and quercitrin. The SPRW increased 5′-nucleotidase with the concomitant decrease of ∆ 5 -3β-HSD, ∆ 5 -17β-HSD and LDH activities in rats exposed in relation to the control. Similarly, the administration of the SPRW caused a systemic oxidative damage along with adverse histopathological changes in germinal epithelial cells. Conclusions: Interestingly, these alterations were differentially reversed by LTME via the elevation of steroidogenic enzymes and cellular ATP.

Background: Toxicities due to fluoride exposure from natural and industrial sources occur commonly in man and animals with severe consequences ranging from mild cardiac derangements to sudden death. In this study, we investigated the protective effects of the methanol extract of Azadirachta indica (AI) against sodium fluoride (NaF)-induced hypertension and genotoxicity in rats. Methods: Sixty rats were divided into six groups of ten rats each as follows: Group A, the control group received distilled water; Group B rats were administered NaF at 600 ppm in drinking water; Groups C and D rats were pre-treated with the methanol extract of AI and thereafter administered NaF at 600 ppm in drinking water for 7 consecutive days; Groups E and F rats were co-administered with AI and NaF. Results: The administration of NaF caused significant (p<0.05) increases in the blood pressure, markers of oxidative stress, serum myeloperoxidase, xanthine oxidase values in NaF-alone treated rats, compared with the control. Significant (p<0.05) decreases were observed in cardiac and renal antioxidant defence system in rats administered NaF alone compared with the control group. NaF treatment also resulted in a reduction in the expressions of extracellular signal-regulated kinase (ERK) 1/2 in cardiac and renal tissues of NaF-treated rats. Moreover, NaF treatment elicited an increase in the frequency of micronucleated polychromatic erythrocytes when compared with the control group. Conclusions: This study shows the protective effect of AI on NaF-induced hypertension and genotoxicity through antioxidant and ERK 1/2 signaling in rats.

Background: Hypoxia inducible factor (HIF)/prolyl hydroxylase domain (PHD)-containing proteins are involved in renal adaptive response to high salt (HS). Peroxisome proliferator activated receptor alpha (PPARα), a transcription factor involved in fatty acid oxidation is implicated in the regulation of renal function. As both HIF-1α/PHD and PPARα contribute to the adaptive changes to altered oxygen tension, this study tested the hypothesis that PHD-induced renal adaptive response to HS is PPARα-dependent. Methods: PPARα wild type (WT) and knock out (KO) mice were fed a low salt (LS) (0.03% NaCl) or a HS (8% NaCl) diet for 8 days and treated with hydralazine. PPARα and heme oxygenase (HO)-1 expression were evaluated in the kidney cortex and medulla. A 24-h urinary volume (UV), sodium excretion (U Na V), and nitrite excretion (U NO x V) were also determined. Results: PHD1 expression was greater in the medulla as compared to the cortex of PPARα WT mice (p<0.05) fed with a LS (0.03% NaCl) diet. The HS diet (8% NaCl) downregulated PHD1 expression in the medulla (p<0.05) but not the cortex of WT mice whereas expression was downregulated in the cortex (p<0.05) and medulla (p<0.05) of KO mice. These changes were accompanied by HS-induced diuresis (p<0.05) and natriuresis (p<0.05) that were greater in WT mice (p<0.05). Similarly, U NO x V, index of renal nitric oxide synthase (NOS) activity or availability and heme oxygenase (HO)-1 expression was greater in WT (p<0.05) but unchanged in KO mice on HS diet. Hydralazine, a PHD inhibitor, did not affect diuresis or natriuresis in LS diet-fed WT or KO mice but both were increased (p<0.05) in HS diet-fed WT mice. Hydralazine also increased U NO x V (p<0.05) with no change in diuresis, natriuresis, or HO-1 expression in KO mice on HS diet. Conclusions: These data suggest that HS-induced PPARα-mediated downregulation of PHD1 is a novel pathway for PHD/HIF-1α transcriptional regulation for adaptive responses to promote renal function via downstream signaling involving NOS and HO.

Background: We investigated the protective effects of amla ( Emblica officinalis ) on the pathogenesis of oxidative stress (OS) and inflammatory response in hypothyroid rats fed with a high-fat diet (HFD) as an experimental model of hypothyroidism (HT) with obesity. Methods: A total of 80 female wistar rats (5-months-old) were divided into eight different groups. Propylthiouracil (PTU) and HFD were used to induce the experimental HT and obesity, respectively. The euthyroid and hypothyroid rats were fed either normal chow or HFD with and without amla extract (AE, 100 mg/kg bw/day) for 6 weeks. The blood and tissues, liver and kidney OS and inflammatory parameters were studied using appropriate biochemical and molecular techniques. Results: PTU and HFD per se caused OS and inflammatory response as evidenced by increased plasma MDA, TNF-α, CRP and GPx in association with decreased levels of TAS and reduced glutathione (GSH). The proteomic analysis revealed that the expressions of pERK, pP38, TNF-α, IL6, COX2 and NOX-4 were up-regulated in the liver and kidney of these rats. In addition, all these metabolic derangements were further augmented when HT was followed by the addition of HFD. This suggested that there was a synergism between HT and the intake of HFD on the development of OS and inflammatory response. Conclusions: The treatment with amla fruit extract significantly restored the redox imbalance and inflammatory signaling and ameliorated OS and inflammatory response, suggesting the use of this natural compound as an alternative remedy or adjuvant for the management of metabolic complications concomitant with HT.

Background: Maternal high fat diet has been implicated in the aetiology of metabolic diseases in their offspring. The hypolipidaemic actions of Cocos nucifera water improve metabolic indices of dams consuming a high fat diet during gestation. This study investigated the effects of C. nucifera water on metabolism of offspring of dams exposed to high fat diet during gestation. Methods: Four groups of pregnant Wistar rat dams (n=6) were treated orally from Gestation Day (GD) 1 to GD 21 as follows: standard rodent feed+10 mL/kg distilled water (Control), standard rodent feed+10 mL/kg C. nucifera water, high fat feed+10 mL/kg distilled water (high fat diet), and high fat feed+10 mL/kg C. nucifera water (high fat diet+ C. nucifera water). The feeds were given ad libitum and all dams received standard rodent feed after parturition. Fasting blood glucose was measured in offspring before being euthanized on Postnatal Day (PND) 120. Serum insulin, leptin, lipid profile and liver enzymes were measured. Results: Serum total cholesterol (TC), insulin, alanine transaminase (ALT) and alkaline phosphatase levels were significantly increased (p<0.05) in high fat diet offspring compared with controls. Similar changes were not observed in high fat diet+ C. nucifera water offspring. Conclusions: Results suggest that the adverse effects of maternal high fat diet on offspring’s metabolism can be ameliorated by C. nucifera water.

Background: Dry eye disease (DED) is a common ocular surface disease significantly affecting the quality of life of patients. The aim of our study is to focus on the prevalence of DED and to determine the relationship between dyslipidemia and DED. Methods: The study was performed with the age group of 25–70 years, who attended the ophthalmology outpatient department at Sri Lakshmi Narayana Institute of Medical Sciences with complaints of dry eye. A standard questionnaire was taken, and tear film tests were performed to diagnose dry eye. Further eyelid margin was examined to detect meibomian gland dysfunction. Based on the tests and examination, patients were grouped as men with and without DED and women with and without DED. Fasting lipid profile was investigated for these groups. Results: The study showed the prevalence of DED mainly in women and found significant association between DED and dyslipidemia. There is a significant relationship between total cholesterol and DED groups especially in women (p<0.001). We also found the association between triglycerides, high density lipoprotein cholesterol, low density lipoprotein cholesterol and DED particularly in women as compared to men. Conclusions: Based on the findings, we emphasize that there is a strong relationship between dyslipidemia and the progression of DED particularly in women. Ophthalmologists may increase their role to educate themselves to diagnose dyslipidemia and ensure comprehensive eye care to prevent blindness and cardiovascular disease. Recent treatment modalities could be aimed to improve the quality of life of women and elderly patients suffering from DED.

Background: Geraniin, a dehydroellagitannin, is a major component of the aqueous extract of the aerial parts of Phyllanthus muellerianus (Kuntze) Exell. ( Euphorbiaceae ). Several Phyllanthus species are traditionally used for painful disorders. The anti-nociceptive effects of the aqueous extract of the aerial parts of P. muellerianus and of geraniin have been scientifically established. The aim of the paper is to determine whether a combination of geraniin and diclofenac or geraniin and morphine leads to better anti-nociceptive effects. Methods: The nature of the interactions of morphine and diclofenac with geraniin was evaluated by undertaking the isobolographic analysis. Mice were treated with geraniin (3–30 mg/kg), morphine (1–10 mg/kg), and diclofenac (10–100 mg/kg) to obtain the ED 50 values of the agents in the formalin test. Dose-response curves were then obtained and analyzed after the co-administration of geraniin with morphine or diclofenac in fixed ratio (1:1) combinations based on specific fractions (1/2, 1/4, and 1/8) of their respective ED 50 values for the formalin test. Results: Geraniin was less potent than morphine but more potent than diclofenac in the formalin-induced nociception. The isobolographic analysis of geraniin/morphine (G/M) and geraniin/diclofenac combinations (G/D) at different fractions revealed the potentiation of their anti-nociceptive effects. The degrees of potentiation, which were calculated as interaction indices, showed synergism for both combinations in both phase I (G/M: 0.040, G/D: 0.017) and phase II (G/M: 0.004, G/D: 0.002) of the formalin test. Conclusions: The present study demonstrates synergism for the co-administration of geraniin with both morphine and diclofenac.

Background: The Icacina senegalensis root bark is traditionally used for the treatment of gastrointestinal disorders in Nigeria. To date, no scientific study has substantiated or refuted this claim. Methods: The antidiarrheal and antimicrobial activities of the ethanol root bark extract were investigated in rats and against some selected diarrhea-causing microorganisms. Results: The extract significantly decreased the frequency of castor oil-induced diarrhea, and inhibited the masses and volumes of intestinal fluid accumulation in the castor oil-induced enteropooling method. The distance travelled by the charcoal meal was also decreased by the extract gastrointestinal transit method. The extract also strongly inhibited the growth of some selected microorganisms. Conclusions: The ethanol extract of the I. senegalensis root bark showed antidiarrheal activity, thus justifying its long folkloric use in diarrhea treatment. The extract also demonstrated antimicrobial activity against selected diarrheal causing organisms.

Background: Irvingia gabonensis stem bark is a medicinal plant used in most parts of Africa to manage a number of ailments including neurodegenerative diseases that occur without scientific basis. This work characterized the phenolic composition, evaluated the cholinergic enzymes (acetylcholinesterase, AChE and butyrylcholinesterase, BChE) inhibition, and assessed the antioxidant activity of phenolic extracts from I. gabonensis (Aubry-Lecomte ex O‘Rorke) Baill bark. Methods: Total phenol and flavonoids content was evaluated in addition to antioxidant activity as shown by Fe 2+ chelation, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability, and 2,2-azino-bis-(3-ethylbenthiazoline-6-sulfonic acid) (ABTS) radical scavenging ability. Inhibitory activities on AChE and BChE were evaluated. Results: The extract was found to be rich in phenolic acid (ellagic acid) and flavonoids (quercetrin, kaempferol, and apigenin). The phenolic extracts displayed DPPH radical scavenging ability (IC 50 =19.98 μg/mL), ABTS radical scavenging ability (IC 50 =18.25 μg/mL), iron chelation (IC 50 =113.10 μg/mL), and reducing power (Fe 3+ to Fe 2+ ) (5.94 mg ascorbic acid equivalent/100 g). Extracts of I. gabonensis inhibited AChE (IC 50 =32.90 μg/mL) and BChE (IC 50 =41.50 μg/mL) activities in concentration-dependent manner. Conclusions: Hence, possible mechanism through which the stem bark executes their anti-Alzheimer’s disease activity might be by inhibiting cholinesterase activities in addition to suppressing oxidative-stress-induced neurodegeneration.
