Cognitive control refers to the internal representation, maintenance, and updating of context information in the service of exerting control over thoughts and behavior. Deficits in cognitive control likely contribute to difficulty in maintaining abstinence in individuals with alcohol use disorders (AUD). In this article, we define three cognitive control processes in detail (response inhibition, distractor interference control, and working memory), review the tasks measuring performance in these areas, and summarize the brain networks involved in carrying out these processes. Next, we review evidence of deficits in these processes in AUD, including both metrics of task performance and functional neuroimaging. Finally, we explore the clinical relevance of these deficits by identifying predictors of clinical outcome and markers that appear to change (improve) with treatment. We observe that individuals with AUD experience deficits in some, but not all, metrics of cognitive control. Deficits in cognitive control may predict clinical outcome in AUD, but more work is necessary to replicate findings. It is likely that performance on tasks requiring cognitive control improves with abstinence, and with some psychosocial and medication treatments. Future work should clarify which aspects of cognitive control are most important to target during treatment of AUD.

To evaluate the contribution of individual synaptic constituents, all available in vivo imaging studies on schizophrenic patients were subjected to a retrospective analysis. For the pool of drug-naïve, drug-free, and acutely medicated patients, major findings were increases in neostriatal dopamine (DA) synthesis and release and decreases in neostriatal DA transporters and D 1 receptors, neostriatal, thalamic, frontal, and parietal D 2 receptors, mesencephalic/pontine and temporal 5-HT 1A receptors, frontal and temporal HT 2A and μ-amino butyric acid (GABA) A receptors. Based on the findings on drug-naïve and drug-free patients, it may be hypothesized that schizophrenia initially is characterized by an impaired mechanism of D 2 autoreceptor and heteroreceptor sensitization leading to sensitization instead of desensitization in response to increased levels of neostriatal DA. Neuroleptic medication blocks neostriatal D 2 autoreceptor and heteroreceptors, reducing neostriatal DA and disinhibiting DA action mediated by D 2 heteroreceptor binding sites. Ultimately, this may result in a restitution of GABA function, leading to a recovery of inhibitory input to the target regions of the descending corticothalamostriatal efferents. Furthermore, a blockade of inhibitory and excitatory neocortical 5-HT function may be inferred, which is likely to reduce (excitatory) DAergic input to the mesolimbic target regions of corticothalamostriatal projections.

Connexins (Cx) play an important role in the coordination of intercellular communication, and autocrine and paracrine regulation of cells within the neurovascular unit (NVU). Gap junctional mechanisms control proliferation and differentiation processes underlying neurogenesis and angiogenesis in the brain. Cx43 possesses some unique properties [the ability to form either intercellular channels permeable for regulatory molecules and ions or hemichannels open to the extracellular space to provide release of cell metabolites; functional coupling with nicotinamide adenine dinucleotide (NAD + )-consuming and NAD + -dependent enzymatic processes] which may be of great importance for the fate of the stem cells. Dynamic changes in Cx43 expression are associated with different stages of brain cells development either at embryonic or adult periods of ontogenesis. This review summarizes recent data on Cx43-controlled neurogenesis in the context of NVU development and functioning. Understanding the molecular mechanisms of gap junctional intercellular communication will support translational studies focused on the development of regeneration-based approaches for the therapy of central nervous system pathology.

Visual cortical areas in the two sides of the brain are interconnected by interhemispheric fibers passing through the splenium of the corpus callosum. In this review, we summarize data concerning the anatomical features of visual callosal connections, their roles in basic visual processing, and how their alterations contribute to visual deficits in different human neuropathologies. Splenial fibers represent a population of excitatory axons with varying diameters, which interconnect cortical columns with similar functional properties (i.e., same orientation selectivity) in the two hemispheres. Their branches activate simultaneously distinct iso-oriented columns in the contralateral hemisphere, thus mediating forms of stimulus-dependent interhemispheric synchronization. Callosal branches also make synapses onto GABAergic cells, resulting in an inhibitory modulation of visual processing that involves both iso-oriented and cross-oriented cortical networks. Interhemispheric inhibition appears to predominate at short latencies following callosal activation, whereas excitation becomes more robust with increasing delays. These callosal effects are dynamically adapted to the incoming visual activity, so that stimuli providing only weak afferent input are facilitated by callosal pathways, whereas strong visual input via the retinogeniculate pathway tends to be offset by transcallosal inhibition. We also review data highlighting the contribution of callosal input activity to maturation of visual function during early ‘critical periods’ in brain development and describe how interhemispheric transfer of visual information is rerouted in cases of callosal agenesis or following splenial damage. Finally, we provide an overview of alterations in splenium anatomy or function that may be at the basis of visual defects in several pathologic conditions.

Febrile seizures (FSs) occur in children older than 1 month and without prior afebrile seizures in the absence of a central nervous system infection or acute electrolyte imbalance. Their pathogenesis is multifactorial. The most relevant familial studies evidence an occurrence rate ranging from 10% to 46% and median recurrence rate of 36% in children with positive familial history for FS. The main twin studies demonstrated a higher concordance rate in monozygotic twins with FS than in dizygotic ones. Linkage studies have proposed 11 chromosomal locations responsible to FS attributed to FEB1 to FEB11. Population-based association studies have shown at least one positive association for 14 of 41 investigated genes with FS. The proinflammatory cytokine interleukin 1β ( IL-1β ) was the most investigated and also gene associated with susceptibility to FS. A possible role in the overlapping of epilepsy and FS was found for 16 of 36 investigated genes. SCN1A , IL-1 β, CHRNA4 , and GABRG2 were the most commonly involved genes in this context. The genetic background of FS involves the regulation of different processes, including individual and familial susceptibility, modulation of immune response, and neuronal excitability and interactions with exogenous agents such as viruses.

Here, we present various research results and thoughts with the intention of challenging notions about free choice. Namely, we describe the concept of transgenerational transmission of epigenetic information and discuss its non-conscious effects on cognitive abilities, behavioral and emotional patterns, and responses with regard to one’s life and decisions, and the impact these have on the concept of free will. In addition, we discuss the essential role of unconscious mechanisms in human decision processes. We also show that twin loss in the womb can have a powerful lifelong impact on the surviving twin through non-conscious context-dependent epigenetic changes. Finally, we hypothesize that human explicit self-consciousness may be an active executer that intermediates between unconsciousness and the external environment by means of feedback and feed-forward interactions. This executive function makes it possible for self-consciousness to continuously develop in self-organized evolution.
