Zusammenfassung Der traditionelle Blick auf die Funktion des Aktin-Zytoskeletts ist in der Regel auf eine Rolle als zytoplasmatisches Zellgerüst eingeschränkt. In den letzten Jahren wurde dieses Konzept dahingehend maßgeblich erweitert, dass dynamische Verän­derungen in der Aktin-Polymerisation ein Signal darstellen, das die Genexpression im Nukleus beeinflusst. Durch diese Kommunikation mit dem Nukleus wird durch zytoplasmatisches und auch nukleäres Aktin eine weitere Ebene der Regulation zellulärer Motilitätsprozesse bereitgestellt. Dabei wurde ein Genregulationskom­plex bestehend aus den Transkriptionsfaktoren SRF (Serum Response Faktor) und Mitgliedern der MRTF-Familie (Myocardin Related Transcription Factor) als primäres Ziel dieser Aktin-Signalgebung in den Nukleus identifiziert. Im Rahmen dieses Übersichtsartikels wird diese aus Aktin-MRTF-SRF bestehende Genregula­tionseinheit in Neuronen vorgestellt.

Zusammenfassung Eines der größten Rätsel der Naturwissenschaften ist der Computer in unserem Kopf: Wie schafft es die Natur, die richtigen Nervenzellen miteinander zu verknüpfen, ohne einem genauen genetisch festgelegten Schaltplan zu folgen? Man nimmt an, dass sich lokale Kontakte im Kortex zunächst zufällig bilden und die ‚falschen‘ im Laufe der Entwicklung wieder entfernt werden. Nur, woher weiß das Gehirn, welche Verbin­dungen ‚falsch‘ und welche ‚richtig‘ sind? Die Mehrzahl der erregenden Synapsen im Gehirn befindet sich auf dendritischen Spines, winzigen pilzförmigen Ausstülpungen der dendritischen Zellmembran. Diese Strukturen faszinieren Neurobiologen seit ihrer Entdeckung durch Ramón y Cajal 1896, da sie festlegen, welche Zellen miteinander Informationen austauschen können. In diesem Artikel geben wir einen Überblick über die zahlreichen Funktionen von Spines und erklären, warum die elektrischen und bio-chemischen Prozesse, die in diesen winzigen Strukturen ablaufen, so wichtig für die Plastizität unseres Gehirns sind.

Zusammenfassung Topografische axonale Projektionen sind ein weit verbreitetes Muster der neuronalen Konnektivität. Das am besten untersuchte Modellsystem für diese Verbindungen ist die retinotektale Projektion. Ihre Entstehung wird im Allgemeinen erklärt durch eine Interaktion gradiert verteilter Marker des ephrin-A/EphA-Systems, welche sowohl auf den Wachstumskegeln retinaler Ganglienzellen als auch auf dem tektalen Zielgebiet exprimiert sind. Die wichtigsten Einsichten in die Entstehung der retinotektalen Projektion lieferten bemerkenswerterweise in vitro- und nicht in vivo-Experimente. So ermöglichten funktionelle in vitro-Assays nicht nur die biochemische Identifikation der postulierten Lenkungsmoleküle, sondern halfen auch, die von ihnen übermittelten Signale zu verstehen. So zeigte sich, dass sowohl forward (ephrin-A -> EphA) als auch reverse signalling (EphA -> ephrin-A) für eine topografiegemäße Lenkung retinaler Axone benötigt werden. Trotz allem existiert im Moment kein Assay, welcher die Topografieentstehung in vitro vollständig reproduzieren kann. Neue in vitro-Techniken wie micro-contact printing oder micro-fluidic networks können dabei helfen, vorhandene Assays weiterzuentwickeln und so ein hinreichendes Set von funktionellen Komponenten der Entstehung topografischer Projektionen zu identifizieren.

Actin, arranged for example in stress fibres, provides a fundamental cytoskeletal frame­work function to all cell types. Notably, there is now mounting evidence that, in addition to cytoplasmic cytoskeletal regulation, ac­tin treadmilling provides a signal modulat­ing nuclear gene expression. In altering gene regulation, cytoplasmic and most likely also a nucleus-resident actin provides an addition­al (gene) regulatory twist to cell motility. So far, the transcription factor serum response factor (SRF) alongside its myocardin-relat­ed transcription factor (MRTF) cofactors has emerged as the main target of actin dynam­ics. In this review, we discuss the impact of actin signalling on nuclear gene expression in the nervous system, where the actin-MRTF-SRF module contributes to various processes including neuronal motility.

One of the biggest remaining mysteries of science is inside our heads: how does nature wire up a high-performance computer with­out having a detailed blueprint specifying the location and strength of every connection? It is assumed that local connectivity in our cor­tex is at first random, and during develop­ment undergoes refinement until only the ‘right’ connections are left over. But how can the brain tell ‘right’ from ‘wrong’ connections? The majority of excitatory connections are formed on dendritic spines, tiny excrescenc­es that cover almost the entire dendritic sur­face of most neurons. Since their discovery by Ramón y Cajal in 1896, neuroscientists have been fascinated by these structures, which ultimately determine which neurons in the brain become connected and form function­al networks. Here we review the many im­portant functions of spines and explain why electrical and biochemical processes in these tiny structures are thought to be crucial for the plasticity of the brain.

Topographic axonal projections are a prev­alent feature of brain connectivity. The ret­inotectal mapping of the chick is the best-studied model system of this type of neuro­nal connectivity. Its formation is common­ly explained by interactions between grad­ed markers of the ephrin-A/EphA family ex­pressed on both retinal ganglion cell growth cones and on the tectal target area. Surpris­ingly, most insights into retinotectal devel­opment have been gathered through in vitro rather than in vivo experiments. In vitro as­says not only enabled the biochemical iden­tification of the postulated molecular mark­ers but also helped to understand the signals conveyed by them. Thus, it was established in vitro that forward (ephrin-A->EphA) as well as reverse signalling (EphA->ephrin-A) are si­multaneously needed for topographically ap­propriate guidance of retinal axons. However, no in vitro assay yet exists that fully reproduc­es topography formation. New in vitro tech­niques such as micro-contact printing or mi­cro-fluidic networks may help to improve ex­istent assays and to identify a sufficient set of functional components that reconstitutes to­pography formation.
