Postmenopausal osteoporosis, a silent epidemic, has become a major health hazard, afflicting about 50% of postmenopausal women worldwide and is thought to be a disease with one of the highest incidences in senile people. It is a chronic, progressive condition associated with micro-architectural deterioration of bone tissue that results in low bone mass, decreased bone strength that predisposes to an increased risk of fracture. Women are more likely to develop osteoporosis than men due to reduction in estrogen during menopause which leads to decline in bone formation and increase in bone resorption activity. Estrogen is able to suppress the production of proinflammatory cytokines like interleukin (IL)-1, IL-6, IL-7 and tumor necrosis factor (TNF-α). This is why these cytokines are elevated in postmenopausal women. In this review article we have made an attempt to collate the various methods and parameters most frequently used for screening of antiosteoporotic activity in postmenopausal osteoporosis. Pertaining to ovariectomized animal model, this is the most appropriate model for studying the efficacy of different drugs to prevent bone loss in postmenopausal osteoporosis.

Background : Medicinal plants have been used for centuries and have become part of complementary medicine worldwide because of their health benefits. Some have been successfully used directly in the treatment and prevention of infectious diseases or indirectly by stimulating the immune system. In the present study, the immunomodulatory activity of the methanol extracts of Adenocarpus mannii, Caucalis melanantha, Ocimum gratissimum, Asystasia intrusa and Clematis chinensis leaves was investigated. Methods : The extracts were prepared by maceration of dry leaves’ powder in methanol. Phytochemical analysis was carried out by chemical reaction methods. The activity of plant extracts was evaluated in in vitro cell cultures by measuring their effect on nitric oxide production by peritoneal macrophages, the proliferation of lymphocytes and the cytotoxic effect on macrophages. The A. mannii extract was further evaluated at 50 and 100 mg/kg body weight in mice for the stimulation of delayed-type hypersensitivity (DTH) reactions and the ability to reverse the myelosuppression induced by cyclophosphamide. Results : All the extract inhibited nitric oxide production by peritoneal macrophages, the highest activity was achieved with C. chinensis extract. However, these extracts did not significantly affect the viability of macrophages. A. mannii, A. intrusa and C. chinensis extracts exhibited stimulatory activity on peripheral blood lymphocytes, whereas C. melanantha and O. gratissimum extracts displayed inhibitory activity. In vivo , the A. mannii extract significantly increased the DTH reaction in mice from 50 mg/kg. This extract also showed a significant increase in the white blood cells and relative weight of the spleen and liver. Conclusions : These results suggest that the A. mannii, C. melanantha, O. gratissimum, A. intrusa and C. chinensis methanol extracts possess immunomodulatory activity. This constitutes additional data on the well-known biological properties of these plants.

Background : Leukocytes play a vital role in the host defence and inflammatory systems, the latter being responsible for the pathogenesis of a wide spectrum of acute and chronic diseases. Green tea is a popular beverage, which is consumed worldwide and its active ingredients are epicatechin derivatives, which possess distinct anti-inflammatory properties. The purpose of this study was to investigate if a green tea extract could enhance leukocyte function in humans. Methods : Volunteers were asked to take 300 mg of the green tea extract daily for 14 days and the capacity of circulating leukocytes to release both myeloperoxidase and lactoferrin was assessed. Whole blood from volunteers was stimulated with the bacterial peptide Formyl-Methionine-Leucine-Phenylalanine (fMet-Leu-Phe). Myeloperoxidase an enzyme that converts hydrogen peroxide to hypochlorous acid and is stored and secreted from the granules of neutrophils and monocytes and was measured as well as lactoferrin which is an iron-binding protein stored and secreted from the neutrophils. In conjunction the antioxidant capacity of the blood of the volunteers was also determined using a chemiluminescence method that measures the capacity of plasma to scavenge superoxide. Results : After 14 days of treatment there was a significant increase in the release of myeloperoxidase and lactoferrin when whole blood was stimulated with fMet-Leu-Phe (p<0.05), which activates a number of leukocytes including mature neutrophils and monocytes. This was mirrored by a significant increase in the total antioxidant status after 14 days of green tea ingestion (p0.05). After the “wash-out” period of 4 weeks, all parameters were consistent with those observed at the start of the trial (day 0). Treatment with the green tea extract also caused a slight but non-significant decrease in the number of circulating leukocytes, but the counts remained within published “normal” ranges for healthy human adults. Conclusions : This study indicates that a green tea extract when taken as a dietary supplement for 14 days can increase the leukocyte activity and the total plasma antioxidant status and may have role to play in the prevention of inflammatory disease.

Background : Newtonia griffoniana (Mimosaceae) is a Central African rain forest tree, whose bark extracts are used in Cameroonian folk medicine for the treatment of anxiety and sleep disorders. Methods : We evaluated the anxiolytic effects of N. griffoniana stem bark methanol extract and its major isolated constituent 2,3,4-trihydroxybutylpentatriacontanoate (newtonoate) on the elevated plus maze. Results : Significant increases in the percentage of entries into open arms were induced by both N. griffoniana extract (100 and 150 mg/kg BW; p<0.01) and newtonoate (doses of 3 and 15 mg/kg BW; p<0.05). Conversely, decreases in the percentage of entries into closed arms were observed at the same doses. In addition, N. griffoniana methanol extract (100 mg/kg) and the isolated newtonoate (30 mg/kg) induced significant (p<0.01 and p<0.05, respectively) increases in the time spent in the open arms, while inducing a decrease in the time spent in the closed arms. Newtonoate treatment also decreased head dipping number at doses of 3 and 15 mg/kg, while N. griffoniana methanol extract induced the same effect at 200 mg/kg. Conclusions : These results suggest that N. griffoniana bark extract has anxiolytic properties, which justify its use in folk medicine. Such effects are at least partly mediated by newtonoate.

Background : Fibrous structures and synthetic polymer blends offer potential usages in making biomedical devices, textiles used in medical practices, food packaging, tissue engineering, environmental applications and biomedical arena. These products are also excellent candidates for building scaffolds to grow stem cells for implantation, to make tissue engineering grafts, to make stents to open up blood vessels caused by atherosclerosis or narrowed by blood clots, for drug delivery systems for micro- to nano-medicines, for transdermal patches, and for healing of wounds and burn care. The current study was designed to evaluate the antimicrobial activity of woven and non-woven forms of nano- and macro-scale blended polymers having biocompatible and biodegradable characteristics. Methods : The antimicrobial activity of non-woven fibrous structures created with the combination of synthetic and biopolymer was assessed using Gram-negative, Gram-positive bacteria, such as Staphylococcus aureus , Proteus vulgaris , Escherichia coli and Enterobacter aerogenes using pour plate method. Structural evaluation of the fabricated samples was performed by Fourier transform infrared spectroscopy. Results : Broad spectrum antibacterial activities were found from the tested materials consisting of polyvinyl alcohol (PVA) with chitosan and nylon-6 combined with chitosan and formic acid. Conclusions : The combination of PVA with chitosan was more bactericidal or bacteriostatic than that of nylon-6 combined with chitosan and formic acid. PVA combination with chitosan appears to be a broad-spectrum antimicrobial agent.

Background : Ethnopharmacological practitioners in Nigeria have used aqueous and ethanol extracts of Bridelia micrantha leaves to manage conditions associated with inflammation, and these include diabetes, chest pain, edema, arthritis and joint pains. This study aimed to evaluate the effects of methanol leaf extract of B. micrantha on chronic inflammation and oxidative stress which accompany diabetic conditions, in streptozotocin-induced diabetic Albino Wistar rats. Methods : The dried leaves were extracted by percolation in 80% methanol:water for 72 h after which the mixture was filtered using Whatman No. 1 (11 μm) filter papers. Acute toxicity studies were done using Wistar rats and given orally up to a dose of 2,000 mg/kg. The animals were monitored for 48 h. The experimental design involved five (5) groups of six (6) albino Wistar diabetic rats each. Groups A, B and C rats received 100, 200 and 400 mg/kg B. micrantha respectively while groups D (negative control) and E (positive control) rats received 10 mL/kg normal saline and 200 mg/kg acetylsalicylic acid (ASA) respectively by gastric gavage for 7 days. Two sterilized cotton pellets (10 mg each) were implanted subcutaneously into both sides of the dorsal area of each diabetic rat in all the groups. Post cotton pellet implantation, rats in three groups (A, B and C) were treated with 100, 200 and 400 mg/kg B. micrantha extract, while those in two groups (D and E) were treated with acetyl salicylic acid (ASA 200 mg/kg) and normal saline (10 mL/kg) respectively by gastric gavage for 7 days. Serum obtained from the animals on Day 8 of the cotton pellet test were used for malondialdehyde (MDA), catalase, superoxide dismutase (SOD) and glutathione (GSH) assays. Results : The administration of the leaf extract up to a dose of 2,000 mg/kg to rats produced absolutely no death or observable signs of toxicity in 48 h. The cotton pellet granuloma weights in 200 mg/kg (44.88±1.2 mg), 400 mg/kg (42.10±1.2 mg) B. micrantha extract treated groups and ASA at 200 mg/kg (43.25±1.8 mg) were significantly lower compared to weight of granuloma (85.50±3.2 mg) obtained in the group treated with normal saline. Serum malondialdehyde (MDA) level in the 200 mg/kg (3.32±0.72 nmol/mL) and 400 mg/kg (1.88±1.27 nmol/mL) B. micrantha extract treated groups were significantly (p<0.05) lower compared to MDA level (6.88±0.79 nmol/mL) in the serum of normal saline treated group. Treatment of diabetic rats with the B. micrantha extract also caused significant (p<0.05) elevation in serum catalase, SOD and GSH levels. Conclusions : The study showed that B. micrantha methanol leaf extract significantly inhibited some chronic inflammation and oxidative stress parameters in diabetes mellitus.

Background : Chromolaena odorata is a plant commonly used traditionally to treat ailments including diarrhoea in Nigeria. Methods : The ethanol leaf extract of C. odorata was studied for its anti-diarrhoeal activity using electrolyte test and castor oil-induced diarrhoea rats’ models. Acute toxicity effect of the extract was also evaluated. Results : The extract showed a dose-dependent protection against castor oil-induced diarrhoea at the tested doses (200 and 400 mg/kg body weights). The protection offered by pretreatment with 400 mg/kg body weight of the ethanol leaf extract of C. odorata with regards to reductions in the incidences of faecal wetness and rate of defaecations were statistically comparable to that achieved with Lomotil, a known anti-diarrhoeic drug. The result of the electrolyte test showed that the extract pretreated groups had significantly (p<0.05) lower potassium and sodium ions in their intestinal fluid when compared with the diarrhoeic untreated controls. This is well tolerated. Conclusions : The results indicate that the ethanol leaf extract of C. odorata is safe and possesses anti-diarrhoeal activity with electrolyte reabsorption proposed as the possible mechanism of action.

Background : Mango ginger ( Curcuma amada Roxb.) is a less-investigated herb for anticancer properties than other related Curcuma species. AKT (a serine/threonine protein kinase B, originally identified as an oncogene in the transforming retrovirus AKT8) plays a central role in the development and promotion of cancer. In this investigation, we have analyzed the effect of supercritical CO2 extract of mango ginger (CA) on the genetic pathways associated with AKT signaling in human glioblastoma cells. Methods : The inhibitory effect of supercritical CO 2 extract of mango ginger ( Curcuma amada ) on AKT signaling was investigated in U-87MG glioblastoma cells. Results : CA was highly cytotoxic to glioblastoma cell line (IC 50 =4.92±0.81 µg/mL) compared to mHypoE-N1 normal mouse hypothalamus cell line (IC 50 =40.57±0.06 µg/mL). CA inhibits AKT (protein Kinase B) and adenosine monophophate -activated protein kinase α (AMPKα) phosphorylation significantly in a dose-dependent manner. The cell migration which is necessary for invasion and metastasis was also inhibited by CA treatment, with about 43% reduction at 20 µg/mL concentration. Analysis of mRNA and protein expression of genes associated with apoptosis, cell proliferation and angiogenesis showed that CA modulates expression of genes associated with apoptosis (Bax, Bcl-2, Bcl-X, BNIP3, caspase-3, mutant p53 and p21), cell proliferation (Ki67) and angiogenesis vascular endothelial growth factor (VEGF). Additionally, heat shock protein 90 (HSP90) and AMPKα genes interacting with the AKT signaling pathway were also downregulated by CA treatment. Conclusions : These results indicate the molecular targets and mechanisms underlying the anticancer effect of CA in human glioblastoma cells.

Background : The objectives of this study were to determine the knowledge and attitude of adult Malaysians about Traditional Chinese Medicine (TCM), to understand the variation in the knowledge and attitude toward TCM among different demographic groups of adult Malaysian population and to determine the pattern of TCM use among adult Malaysians. Methods : This study was a descriptive, cross-sectional survey using the convenience sampling method. A total of 400 adult Malaysians were recruited for this study. An interview-administered questionnaire was used. Descriptive statistics Mann-Whitney U Test and Kruskal–Wallis H test were used to analyze the data. The reliability of survey data in this study may affect since closed-ended questions are used. Results : The more commonly utilized therapy was herbal medicine (n=192, 82.1%), acupuncture (n=79, 33.8%) and oriental massage (n=58, 24.8%). The mean knowledge score and mean attitude score for the respondents were 5.00±1.71 and 7.17±2.10 respectively. There was a significant difference in the mean knowledge score between genders, among TCM users and non-TCM users, people of different education levels and ethnicity. Conclusions : There were more TCM users than non-TCM users and the use of TCM was higher in females than males. The most popularly used TCM modalities were herbal medicines and acupuncture. Gender, education, ethnicity and use of TCM had a significant effect on adults’ knowledge toward TCM.

Background : To manage chronic constipation, numerous lifestyle modification schemes and recommendations as well as applications of natural medicaments have been mentioned in manuscripts of traditional Persian medicine (TPM). This study was aimed to compare the impacts of some of those recommendations with lactulose, on functional chronic constipation. Methods : Via a blocked randomization, 100 patients were enrolled. Schemes and recommendations from TPM as intervention group were evaluated versus lactulose as control by weekly follow-ups with standard questionnaire for 3 months. Stool frequency, hard stool, painful defecation, incomplete evacuation sensation, anorectal obstruction sensation and manual maneuvers were considered as outcome measures. Results : Eighty-six patients (42 in schemes and 44 in lactulose groups) completed the study. Median weekly stool frequency in 0, 4, 8 and 12 weeks of treatment was 1.76±1.79, 2.88±0.89, 2.95±1.05 and 2.93±1.11 in the schemes and 2.41±1.67, 2.57±0.90, 2.84±0.91 and 2.77±1.00 in lactulose groups, respectively (p=0.10, 0.11, 0.60, 0.51). Thirty-two (76.2%) patients in schemes and 24 (54.5%) patients in lactulose groups were treated at the end of the protocol as they did not meet the Rome III criteria for constipation (p=0.04). In schemes group, patients reported no undesirable effects, whereas seven (15.9%) in lactulose group reported flatulence (p=0.02). Conclusions : Studied schemes were as effective as lactulose, a gold standard to manage constipation. Results demonstrated that TPM schemes and recommendations, as lifestyle modification, for at least 3 months can be introduced as cheap, available and accessible approaches for the management of constipation.
