Micelles Activated Planar Chromatographic Separation of Hydrophilic Vitamins

Abstract

The separation efficiency of micellar solutions in thin layer chromatography of hydrophilic vitamins (B1, B9, B12 and C) has been examined using silica gel as stationary phase. The best results were obtained with mixed micelles (aqueous solution of anionic SDS (0.5%) plus non-ionic Triton X-100 (0.5%) surfactants) and butanol (9:1 v/v) as mobile phase. The proposed method is applicable to the identification and separation of vitamins (B1, B9 and C) present in drug samples (Becosule and Celin). The separation 1 μg of vitamin C from milligram quantities (up to 0.50 mg) of vitamin B1 has been successfully realized. Effect of metal ions as impurities on the separation of B1, B9 and vitamin C and limit of detection of these vitamins have been examined.

Kurzfassung

Die Trennungseffizienz mizellarer Lösungen bei der Dünnschichtchromatographie von hydrophilen Vitaminen (B1, B9, B12 und C) wurde unter Verwendung von Silca-Gel als stationäre Phase geprüft. Die besten Resultate wurden mit gemischten Mizellen (wässrige Lösung von anionischem SDS (0,5%) plus nichtionischem Triton X-100 (0,5%) Tensiden) und Butanol (9:1 v/v) als mobile Phase erreicht. Die vorgeschlagene Methode ist geeignet zur Identifikation und Trennung von Vitaminen (B1, B9 und C), die in Medikamentenproben (Becosule und Celin) enthalten sind. Die Trennung von 1 μg Vitamin C aus Milligrammengen (bis zu 0,50 mg) an Vitamin B1 konnte erfolgreich realisiert werden. Der Einfluss von Metallionen als Verunreinigungen auf die Trennung von B1, B9 und Vitamin C sowie die Detektionsgrenze dieser Vitamine wurde geprüft.

Ali Mohammad is Senior Faculty in the Department of Applied Chemistry, Faculty of Engineering and Technology, Aligarh Muslim University, Aligarh, India. His scientific interests include physicoanalytical aspects of solid state reactions, micellar thin layer chromatography and surfactants analysis. He has supervised 30 students for Ph.D./M.Phil degrees. He is author or coauthor of over 175 scientific papers and book chapters in the area of chromatography. He has authored (and then updated) the Chapter on “TLC of Inorganics and Organometallics” for all three editions of the Handbook of Thin-Layer Chromatography edited by Professor J. Sherma and B. Fried (published by Marcel Dekker). Dr. Mohammad is serving as editor of Journal, Chemical and Environmental Research being published from India since 1992, covering all aspects of Environmental Sciences. He is also the Associate Editor for Analytical Chemistry section of Journal of Indian Chemical Society. He is currently serving on the editorial boards of Acta Chromatographica, Acta Universitatis Cibiniensis Seria F. Chemia and Annals of Agrarian Science. Dr. Mohammad obtained his M.Phil (1975), Ph.D. (1978) and D.Sc. (1996) degrees from Aligarh Muslim University, Aligarh, India.

Miss Ruby Guptai is currently working as research scholar in the department of Applied Chemistry. She received her M.Phil. degree from Aligarh Muslim University, Aligarh, India in 2006. She has published four research papers.

Mr. Showkat Ahmad Bhawani is currently working as research scholar in the department of Applied Chemistry. He received his M.Sc. and M.Phil. degrees from Aligarh Muslim University, Aligarh, India in 2003 and 2006, respectively. He has published six research papers.
